摘要
目的 观察脊髓损伤后雪旺细胞移植对运动终板的保护作用。方法 培养纯化并S100鉴定雪旺细胞。70只Wistar大鼠分别分为对照组、单纯损伤组、损伤加移植细胞组,移植细胞时间分别为伤后1小时、4小时、8小时、12小时、24小时,每组分别伤后1周、4周及8周取材。评定细胞移植后存活率及测定脊髓组织LDH值,并对腓肠肌运动终板行ACh E和CGRP免疫组化染色,测定表达量。结果 雪旺细胞第4代时纯度为95.3%,移植不同时间存活率无统计学差异(P〉0.05),早期移植雪旺细胞LDH值有明显下降并且伤后8~12小时为早期时间窗,损伤加移植细胞组相比单纯损伤组在伤后1周、4周及8周ACh E及CGSP组织化学显示运动终板染色较深(P〈0.05)。结论 雪旺细胞早期移植可以保护脊髓损伤后运动终板,可能通过减少LDH的释放起作用。
Objective To observe protection effect of Schwann cells to motor end plate after spinal cord injury. Methods Sehwann cells were cultured, purified and identified with S100. Seventy Wistar rats were divided into control group, damage group, damage and cell transplantation group. The time points of cell transplantation were 1 hour, 4 hours ,8 hours, 12 hours, 24 hours after injury, respectively. Specimens were taken 1 week, 4 weeks and 8 weeks after injury from each group. Then we evaluated survival rate of transplanted cells and determinate LDH values of the spinal cord tissue. AChE and CGRP immunohistochemical staining of the motor end plate of the gastrocnemius muscle was performed to measure their expression. Results The purity of Schwann cells of the 4th generation was 95.3%, and transplant survival rate for different time showed no statistical difference ( P 〉 0. 05 ). LDH values declined obviously when transplantation of Sehwann cells at early stage and 8 to 12 hours after injury can be seen as early time window. Injury and cell transplantation group compared with the only damage group in 1 week, 4 weeks and 8 weeks after injury had deeper dye in motor endplate by doing AChE and CGSP histochemistry (P 〈 0. 05). Conclusions Early transplantion of schwann cells can protect the motor end plate after spinal cord injury, which may work by reducing the release of LDH.
