亲和色谱应用于天然活性成分筛选的研究进展 被引量:12

Progresses in screening active compounds from herbal medicine by affinity chromatography

摘要 亲和色谱法(affinity chromatography)是一种利用固定相的结合特性来分离分子的色谱方法。利用化合物和固定相表面之间的某种特异性亲和力,将目标化合物从大量无亲和活性的化合物中分离出来。该方法可同时进行色谱分离和活性筛选,作为一种高通量筛选方法广泛应用于各种活性药物的筛选。该文主要针对亲和色谱的发展历史、筛选技术类型及其在天然活性成分筛选中的应用进行了综述。并就其应用前景进行讨论,以期更好地发挥亲和色谱在药物筛选中的应用。 Affinity chromatography is a chromatographic method for separating molecules using the binding characteristics of the stationary phase with potential drug molecules. This method can be performed as a high throughput screening method and a chromatographic separation method to screen a variety of active drugs. This paper summarizes the history of affinity chromatography,screening technology of affinity chromatography,and application of affinity chromatography in screening bio-active compounds in herbal medicines,and then discusses its application prospects,in order to broaden applications of the affinity chromatography in drug screening.
