荧光PCR和脉冲场电泳技术在一起霍乱弧菌疫情诊断中的应用

摘要 目的探讨荧光PCR和脉冲场电泳(PFGE)在霍乱弧菌疫情调查中的应用,为霍乱防治提供依据。方法采集疫情相关标本,进行分离培养鉴定和荧光PCR检测,霍乱弧菌分离菌株基因组进Not I酶切,通过PFGE获得电泳图谱,利用BioNumerics软件对图谱进行聚类分析。结果血清分型分离菌株为霍乱弧菌0139群,标本荧光PCR检测结果毒力基因呈阳性,该起疫情分离的8株霍乱弧菌毒株PFGE图谱一致。结论荧光PCR可快速准确鉴定病原体,PFGE具有较强的菌株同源性分析能力,两种方式的联合应用能在突发公共卫生事件的快速诊断及疫情处置中发挥重要作用。
