DPP-4抑制剂对糖尿病大鼠血管内皮细胞VCAM-1及eNOS表达的影响 被引量:2

The effect of DPP-4 inhibitor on VCAM-1 and eNOS of vascular endothelial cell in diabetes rats

摘要 目的:观察DPP-4抑制剂磷酸西格列汀对1型糖尿病大鼠血管内皮细胞eNOS及VCAM表达的影响。方法:28只SD大鼠,分为对照组8只;剩余20只注射STZ(50mg/kg),1周后根据血糖标准分为模型组6只,干预组7只,模型组和对照组给予生理盐水,干预组给予磷酸西格列汀,总共灌胃5周。每周检测大鼠体重和血糖,干预第5周取组织用RT-PCR检测eNOS,VCAM-1的表达。结果:STZ注射后,大鼠血糖升高,西格列汀干预后,与干预前相比,血糖水平无明显变化。西格列汀干预后,与模型组相比,大鼠eNOS表达升高,VCAM-1表达下降(P<0.01)。结论:磷酸西格列汀可以促进1型糖尿病大鼠血管内皮细胞eNOS表达,抑制VCAM-1的表达,从而可能改善糖尿病大鼠血管内皮细胞功能。 Objective:To observe the DPP-4inhibitor Sitagliptin on vascular endothelial cell in diabetes rats.Method:Twenty-eight rats were divided into control group(n=8)and the Streptozotocin injection group(STZ,50mg/kg,n=20).One week later,the rats were randomly divided into model group(n=6)and treatment group(n=7)according to the standard of blood glucose.The control and model groups were received physiologic saline,and the treatment groups were administered with sitagliptin for 5weeks.The blood glucose and the weight of rats in both group were detected each week,and the expression of eNOS,VCAM-1were measured using RT-PCR after 5weeks treatment.Result:The blood glucose level increased after injected STZ.But there were no significant changes for the level of blood glucose after treatment with Sitagliptin.Compared with the model group,the expression of eNOS increased,VCAM-1decreased in treatment group(P〈0.01).Conclusion:Sitagliptin can enhance the expression of eNOS,inhibit the expression of VCAM-1,probably through improving vascular endothelial cell function of diabetes rat.
