摘要
目的探讨小鼠眼球标本石蜡切片的制作方法。方法处死C57BL/6小鼠,取眼球固定于质量分数为4%中性多聚甲醛中,通过严格控制脱水与浸蜡程序并制成石蜡切片,经HE染色后在显微镜下观察。结果眼球外形均无明显变形和收缩;显微镜下视网膜组织结构完整清晰,各层细胞排列紧密,无明显裂隙。结论该方法适合小鼠眼球标本切片,尤其是视网膜切片的一种有效制作方法。
Objective To determine optimal HE staining procedures on paraffin sections of mice eyeball.MethodsThe C57BL/6mice were killed by using CO2.The eyeballs of the mice were taken and immersed into 4% neutral paraformaldehyde（PFA）solution.The fixed eyeballs were dehydrated and embedded in paraffin.The eyeballs were cut to 5um thickness sections.After HE staining,the sections were observed under a light microscope.ResultsThere was no obvious eyeball distortion or retinal detachment.The structure of retinalthe eyeballs was clear and intact.The cells in retinal were arranged regularly and compactly.Conclusion The improved method for paraffin sections is more effective than others and fit to paraffinsections of mouse eyeball.
