摘要
目的:探讨子宫颈病变中Aurora-A激酶(Aurora-A)、微小染色体维持蛋白7(MCM7)及人乳头瘤病毒16型E7蛋白(HPV16 E7)的表达及三者相关性。方法半定量反转录-聚合酶链反应(RT-PCR)法检测20例低级别子宫颈上皮内瘤变( CINⅠ)、30例高级别子宫颈上皮内瘤变( CINⅡ-Ⅲ)、40例子宫颈鳞状细胞浸润癌及20例慢性子宫颈炎组织中Aurora-A、MCM7及HPV16E7 mRNA的表达。结果①Aurora-A mRNA在CINⅡ-Ⅲ组、子宫颈癌组的表达分别高于慢性子宫颈炎组、CINⅠ组,差异有统计学意义(P<0.05),CINⅡ-Ⅲ组与子宫颈癌组比较,差异无统计学意义(P>0.05)。②MCM7 mRNA表达从慢性炎症到浸润癌呈渐进性增高,慢性子宫颈炎组、CINⅠ组分别与CINⅡ-Ⅲ组、子宫颈癌组比较,差异有统计学意义(P<0.05),CINⅡ-Ⅲ组与子宫颈癌组比较,差异有统计学意义(P<0.05),慢性子宫颈炎组与CINⅠ组比较,差异无统计学意义(P>0.05)。③HPV16E7 mRNA的表达在子宫颈癌组和CINⅡ-Ⅲ组显著高于慢性子宫颈炎组及CINⅠ组(P<0.05);而子宫颈癌组与CINⅡ-Ⅲ组比较,差异无统计学意义( P>0.05)。④在 CIN 中, Aurora -A、MCM7 mRNA 的表达在HPV16E7阳性组高于阴性组,差异均有统计学意义(P<0.05)。在浸润癌中,Aurora-A与HPV16E7的表达呈正相关(r=0.642,P<0.05),MCM7与HPV16 E7的表达呈正相关(r=0.608,P<0.05),Aurora-A与MCM7的表达呈正相关(r=0.734,P<0.01)。结论 Aurora-A、MCM7及HPV16E7 mRNA的表达随子宫颈病变进展逐渐增加,子宫颈癌中三者表达呈正相关;Aurora-A、MCM7及HPV16E7与子宫颈癌的发生、发展密切相关,检测子宫颈组织中三者表达,可用于子宫颈高级别CIN和子宫颈癌的筛查或预测。
Objective To explore the relationship among Aurora -A, minichromosome maintenance protein 7 (MCM7) and human papillomavirus 16 E7 (HPV16E7) according to their expression in cervical intraepithelial neoplasia (CIN) and cervical invasive squamous cell carcinoma (SCC).Methods The expressions of Aurora -A, MCM7 and HPV16E7 were measured by reverse transcription -polymerase chain reaction ( RT-PCR) using sample tissues collected from twenty patients with low -grade cervical intraepithelial neoplasia ( CIN I) , thirty with high-grade cervical intraepi-thelial neoplasia ( CIN II-III) , forty with cervical invasive squamous cell carcinoma ( SCC) and twenty with chronic cer-vicitis.Results The expression of Aurora-A mRNA was significantly higher in the CIN II -III and cervical invasive SCC groups than either the chronic cervicitis or CIN I group ( P〈0.05) .No significant difference was seen between the CIN II-III and cervical invasive SCC group (P〉0.05).The expression of MCM7 mRNA was progressively enhanced from tissues of chronic cervicitis to cervical invasive SCC .Its expression was dramatically strengthened in the CIN II -III and cervical invasive SCC group , compared with the chronic cervicitis and CIN I groups , respectively (P〈0.05), while no significant difference was seen between the CINⅡ-Ⅲand cervical invasive SCC groups (P〈0.05), as well as the chronic cervicitis and CIN I groups (P〉0.05).The expression of HPV16E7 mRNA was remarkably increased in the CIN II-III and cervical invasive SCC groups , in comparison with those in either the chronic cervicitis or CIN I group ( P〈0.05), while no statistical difference was seen between the CIN II -III and cervical invasive SCC groups (P〉0.05). The CIN patients with HPV16E7 presented significantly higher amounts of Aurora -A and MCM7 mRNA than the nega-tive carriers (P〈0.05).Using invasive cervical cancer tissues , positive correlations were determined between the ex-pressions of Aurora -A and HPV16E7 (r=0.615, P〈0.05), MCM7 and HPV16E7 (r=0.608, P〈0.05) as well as Aurora-A and MCM7 (r=0.734, P〈0.01).Conclusion The mRNA levels of Aurora -A,MCM 7 and HPV16E7 are progressively increased with the pathological changes of the cervix , which are positively correlated in cervical cancer tissues.These findings indicate potential roles of these proteins in the pathogenesis of cervical cancer .Their levels can be measured for screening and predicting high -grade CIN and SCC .
