育发剂对中国仓鼠肺细胞活力和染色体畸变的影响研究

Influence of Hair Restorer on Cell Viability and Chromosomal Aberration of CHL Cell

摘要 目的评估2种育发剂对中国仓鼠肺细胞(CHL)细胞活力的影响及其诱导染色体畸变的能力。方法在非代谢活化和代谢活化情况下,采用MTT法和体外哺乳动物细胞染色体畸变试验检测2种(编号A和B)育发剂对CHL细胞活力和染色体畸变率的影响。结果 A育发香波在不含S9的系统中,受试物终浓度为0.156μl/ml时细胞存活率为54.9%;在含S9的系统中,受试物终浓度为0.313μl/ml时细胞存活率为52.4%;B育发膏在不含或含S9的系统中,受试物终浓度为5 mg/ml时细胞存活率均超过90%。与对照组比较,A育发香波各剂量组的染色体畸变率差异无统计学意义(不加S9的体系下Pearson Chi-Square值=0.341,P=0.952,在S9体系下的Pearson Chi-Square值=0.687,P=0.876),B育发膏在加S9和不加S9试验体系下高剂量组的染色体畸变率与阴性对照组相比,有显著性差异(不加S9的体系下Pearson Chi-Square值=16.611,P=0.000,在S9体系下的Pearson Chi-Square值=11.971,P=0.01;重复试验不加S9的体系下Pearson Chi-Square值=15.457,P=0.000,在S9体系下的Pearson Chi-Square值=15.341,P=0.000),低、中剂量组的染色体畸变率无显著性差异(不加S9的体系下Pearson Chi-Square值=0.000,P=1.000,在S9体系下的Pearson Chi-Square值=0.687,P=0.709)。结论在本实验条件下,A育发香波无致突变性,B育发膏有致突变性。 Objective To evaluate the influence of two kinds of hair restorer on cell viability and chromosomal ab- errations of Chinese hamster lung (CHL) cells. Methods CelI viability was assessed by MTr assay and chro- mosome damage was detected by in vitro mammalian cells,: chromosome aberration test in the presence and ab- sence of the liver homogenate fraction ( $9 mix). Results Cell viability treated with A Shampoo was 54. 9% when A shampoo at 0. 15625 uL/mL in the absence of $9 mix, while it was 52. 4% when A Shampoo at 0. 3125 uL/mL in the presence of S9 mix. Cell viability treated with B ointment was over than 90% when B ointment at 5 mg/mL both in the presence and absence of,S9 mix. Chromosomal aberration rate in A Shampoo treated group showed no statistical significance withH'20 hegative control group with or without S9 mix (With- out $9 mix: Pearson Chi - Square value =0. 341, P =0. 952, With S9 mix: Pearson Chi - Square value = 0. 687, P = 0. 876). Chromosomal aberration rate in high - dosage B ointment treated group was significantlydifferent from solvent control group (without S9 mix: Pearson Chi - Square value = 16. 611, P = 0. 000, with s9 mix: Pearson Chi - Square value = 11. 971, P = 0. 01; Repeat Test without S9 mix: Pearson Chi -Square value = 15.457, P = 0. 000, with S9 mix : Pearson Chi - Square value = 15.341, P = 0. 000). There were no significant differences in terms of chromosome aberration rate between low - dosage and middle - dosage B Ointment groups and solvent control group (without S9mix: Pearson Chi - Square 1. 000, with S9 mix: Pearson Chi - Square value = 0. 687, P = 0. 709 ). Conclusion A shampoo shows no mutagenicity on CHL cell, while B ointment shows mutagenicity on on CHL ceils. value = 0. 000, P = Under this condition, chromosome aberration
