摘要
探讨了龙须草叶片扫描电镜样品的制备技术以及扫描电镜下电压的选择。结果表明:取叶组织直接镀金观察的效果与通过戊二醛固定经丙醇脱水后再镀金观察的效果相差不大,研究龙须草叶片结构可以用直接制片法。为了获得清晰的龙须草叶片扫描电镜图,在扫描电镜下选择用10 k V的电压为最佳。
The SEM sample preparation technique of Eulaliopsis binata blade and scanning electron microscope voltage selection were studied. The results showed: the observation effect of blade tissue by directly gold plating was not very different from blade tissue by glutaraldehyde fixing and alcohol dehydration and gold plating,most of the blade structure studing can be used to the method of directly gold plating. The 10 KV was the best choice to observe blade structure which can get more clear picture.
