兴义鸭MEF2A基因的SNPs筛选及其生物信息学分析 被引量:2

摘要 为了解鸭MEF2A基因的功能和结构以及其对鸭遗传效应的影响,采用混合DNA池、基因克隆、PCR产物直接测序和生物信息学相结合的方法 ,对兴义鸭的MEF2A基因进行研究。结果并未发现SNP位点,运用生物信息学方法对其蛋白质一级结构和二级结构进行分析,发现其包含33个α螺旋、20个β折叠、44个无规则卷曲。将获得的鸭的MEF2A蛋白序列与NCBI上提交的家鼠、牛、斑马鱼、猪、鸡的MEF2A序列相比对,结果显示:鸭与鸡、鱼的亲缘关系相近,与家鼠的亲缘关系较远。 In order to understand the structure and function of duck MEF2 A gene and to understand their genetic effect on duck, MEF2 A gene was cloned by bioinformatics and PCR, and the polymorphisms of exon of MEF2 A gene in Xingyi duck were detected by direct sequencing of PCR products. The results showed that there was no SNPs. Using bioinformatics method, the primary protein structure and secondary structures were analyzed, the results showed that, the protein contained 33 α-helics, 20 β-sheets and 44 irregular coils. We got the duck MEF2 A protein sequences and blasted MEF2 A protein sequences to Mus musculus, Bos tarurs, zebra fish, Susscrofa domestica, Gallus domestiaus in NCBI, the results showed that duck had nearly genetic relationship with G.domestiaus and zebra fish, had far genetic relationship with M. musculus.
