隐丹参酮对乳腺癌MDA-MB-231细胞自噬和侵袭迁移的影响 被引量:11

摘要 目的:研究隐丹参酮对乳腺癌MDA-MB-231细胞自噬、增殖和侵袭迁移的作用,初步探讨其抗肿瘤的作用机制。方法:用不同浓度隐丹参酮处理MDA-MB-231细胞24,48,72h,MTT法检测细胞活性,计算24h半数抑制浓度(IC50);20μmol/L隐丹参酮处理细胞24h,吖啶橙染色检测细胞酸性自噬囊泡;Western blot检测自噬标志蛋白LC3-Ⅱ、LC3-Ⅰ(微管相关蛋白1轻链3)及Beclin 1的表达;Transwell小室观察细胞侵袭力;划痕实验检测细胞迁移力。结果:隐丹参酮呈时间-浓度依赖性抑制MDA-MB-231细胞增殖(P<0.05);与未加药组相比,隐丹参酮处理组细胞酸性自噬囊泡增多,Beclin 1蛋白表达增多且LC3Ⅱ/LC3Ⅰ值增高(P<0.05);transwll小室及划痕实验发现,经隐丹参酮处理的MDA-MB-231细胞穿膜数及迁移数均较未加药组少,差异具有统计学意义(P<0.05)。结论:隐丹参酮可诱导乳腺癌MDA-MB-231细胞自噬,从而抑制其增殖和侵袭迁移的潜能。
