升红颗粒的提取纯化工艺研究

摘要 目的:优选升红颗粒提取纯化工艺的最佳条件。方法:处方中的鸡血藤和花生红衣用水提取,提取液浓缩后醇沉纯化。设计正交试验方法,以儿茶素、总黄酮含量为评价指标,对水提取工艺条件进行考察;以儿茶素、总黄酮转移率和浸膏得率为指标对醇沉纯化工艺条件进行考察。结果:水提取的最佳工艺条件为加水量为药材量的12倍,每次提取时间为1.5h,共提取3次;醇沉纯化的最佳工艺条件为浓缩液相对密度为1.10~1.15g/ml (30℃),含醇量为70%,静置时间36h。结论:优选出的最佳工艺条件经验证其结果稳定,指标性成分含量高,可以用于投入生产。 Objectice To optimize the conditions of the extraction and purification technique for Shenghong granules.Methods Herb Caulis Spatholobi and peanut coat in formulation were extracted by water decocting,and the condensed extracted liquid was puri-fied by ethanol precipitating.Taking the content of catechin and total flavonoids as evaluation markers for the extraction technique of water decocting,and the transferring rate of the catechin and total flavonoids,and extract yield as evaluation markers for the purifica-tion technique of ethanol precipitating,each technique condition was evaluated by orthogonal design.Results The optimum condition of water decocting process was that the crude herb powder were decocted for 3 times with 12-fold water,1. 5 hour each time.The op-timum condition of ethanol precipitating process was that extracted liquid was precipitated 36 hours,which relative density was 1. 10-1. 15g/ml (30℃)and concentration of ethanol was 70% .Conlusions The repeated experimental results are stable on the optimum conditions,and the contents of components for markers are high.which could be used for enlarged production.
