摘要
为观察中药地翁颗粒在体外对小鼠脾淋巴细胞功能的影响,设置高(100mg/L)、中(10mg/L)、低(1mg/L)3个剂量组,采用3 H法进行刀豆蛋白A(ConA)、脂多糖(LPS)诱导脾淋巴细胞增殖试验,ELISA方法检测ConA或金黄色葡萄球菌Cown株灭活菌体(SAC)刺激的脾淋巴细胞上清液IL-2、IL-6、IL-10、IL-12、IFN-γ和TNF-α的含量。结果显示,在安全浓度范围内,地翁颗粒能够明显促进T淋巴细胞和B淋巴细胞增殖;能够促进ConA刺激的淋巴细胞分泌INF-γ;能够促进SAC刺激的淋巴细胞分泌IL-12、INF-γ和IL-6,并抑制IL-10的分泌。结果表明,地翁颗粒能够有效调节小鼠脾淋巴细胞的功能,促进细胞免疫。
To examine the effect of Diweng Keli on immune function of mouse spleen lymphocyte in vitro,the blastogenesis of the responder cells was quantified by a colorimetric method using MTT in 96-well microculture plates. Then,we investigated the effects of Diweng Keli on the proliferative activity of lymphocytes stimulated by concanavalin A(Con A) and lipopolysaccharide(LPS) by 3 H- thymidine and cytokine levels in the supernatant by ELISA. The results showed that Diweng Keli promotes the proliferative activity of lymphocytes stimulated by concanavalin A(Con A)and li- popolysaccharide(LPS) ,the production of IFN-)' of T cells and the production of IL-12,IFN-7 and IL-6 of antigen-presenting cells(APC) ,but inhibits the production of IL-10 of APC. These results indicated that Diweng Keli promotes cell-mediated immunity of mice and regulate mice immune function.
