摘要
目的:建立测定人血浆依托咪酯浓度的HPLC方法。方法色谱柱为AgilentTC-C18柱(250mm×4.6mm,5μm),流动相为甲醇-水(75:25),流速为1.0mL/min,检测波长为241nm。结果依托咪酯浓度在0.25~5.00μg/mL范围内与峰面积呈良好线性关系(r=0.9987),方法回收率在98.6%~101.3%,提取回收率在72.6%~74.6%。结论本方法操作简便、准确、可靠,适用于测定人血浆中依托咪酯的浓度。
Objective To develop a method for the determination of etomidate concentration in plasma by using HPLC and to provide scientific analysis for the studies of etomidate clinical pharmacology. Methods The Agilent TC-C18 (250mm×4.6mm,5μm) was used as analytical columu with mobile phase consisted of methanol and water (75:25) .The flow rate was 1.0 mL/min and detection wavelength was 241nm. Results The linear ranges were 0.25-5.00μg/mL for etomidate in human plasma.The method recover were 98.6%-101.3%and the extraction recovery were 72.6%-74.6%. Conclusion The method is simple, accurate and reliable which is suitable for the determination of etomidate in human plasma.
