摘要
目的探讨结肠癌转移相关基因1(MACC1)在食管癌细胞株Eca-109中的作用及其可能机制。方法体外培养Eca-109细胞,并分为MACC1shRNA转染重组质粒组(A组)、转染空质粒阴性对照组(B组)和Eca-109细胞空白组(C组)。采用RT-PCR和Western blot法分别检测MACC1、c-MET mRNA和蛋白表达,Transwell和划痕实验分别观察转染前后Eca-109细胞侵袭和迁移能力的变化。结果与B、C组相比,A组MACC1、c-MET mRNA和蛋白表达下调,Eca-109细胞侵袭和迁移能力降低(P<0.05)。结论下调MACC1表达能明显抑制食管癌细胞的侵袭和转移能力,可能是通过调节下游基因c-MET表达实现的。
Objective To investigate the role and underlying mechanism of metastasisassociated colon cancer 1(MACC1)in esophageal carcinoma cell line Eca-109.Methods Eca-109 cells were cultured in vitro and divided into three groups of A(transfected with recombinant plasmid p-super-EGFP-MACC1shRNA),B(transfected with empty plasmid p-super-EGFP-1)and C(blank control).The mRNA and protein expressions of MACC1 and c-MET were detected by RT-PCR and Western blot,respectively.The invasion and migration of Eca-109 cells were measured by Transwell and wound-healing assays.Results Compared with groups of B and C,the mRNA and protein expressions of MACC1 and c-MET were downregulated,and the invasion and migration of Eca-109 cells were decreased in group A(P〈0.05).Conclusion Downregulation of MACC1 expression can significantly inhibit the invasion and migration of esophageal carcinoma cells probably via downregulating the expression of c-MET.
