Genotype features of human astrovirus in Xuzhou area in 2013

摘要 目的 了解徐州地区腹泻患者中人星状病毒(HAstV)的主要基因型别和流行病学特征.方法 收集2013年江苏省徐州市疑似病毒性腹泻患者粪便标本358份,采用荧光定量PCR方法检测粪便中星状病毒RNA,并对部分阳性标本双区测序以明确基因型.计数资料比较采用x2检验.结果 358份粪便标本中,共检出HAstV阳性标本25份,占6.9%.HAstV感染的高峰季节在7至9月份,各年龄段人群均易感染,0~5岁人群感染率较高.对部分HAstV阳性标本的测序结果显示,开放读码框(ORF) la区测序的21份标本中11株为HAstY-1型,2株为HAstV-2型,4株为HAstY-8型,4株为HAstV-3型;ORF 2区测序的16份标本中10株为HAstV-1型,2株为HAstV-2型,4株为HAstV-5型,未检出其他型别.其中HAstV-1型存在两种不同的亚型,分别与上海株(FJ375759)及俄罗斯株(JF929185)有较高同源性.结论 HAstV可能是徐州地区病毒性腹泻的重要病原体,以HAstV-1型为主,同时也存在其他型别的感染. Objective To investigate the prevalent genotype and epidemiological characteristics of human astrovirus (HAstV) in patients with diarrhea in Xuzhou area.Methods A total of 358 fecal specimens were collected from patients with viral diarrhea at Xuzhou in 2013.HAstV RNA was detected by fluorescence quantitative polymerase chain reaction,and genotypes were determined by sequence analysis in some positive specimens.Enumeration data were analyzed using chi-square test.Results Among all the 358 samples,25(6.9%) eases were positive for HAstV.The HAstV infections peaked from July to September.Patients of all ages were susceptible to HAstY,especially in those 0-5 years old.Sequence analysis in some positive specimens showed that,among all 21 specimens sequenced at region open reading frame (ORF)1a,11 HAstV-1 strains,2 HAstV-2 strains,4 HAstV-8 strains and 4 HAstY-3 strains were found.Among 16 specimens sequenced at region ORF 2,10 HAstV-1 strains,2 HAstV-2 strains and 4 HAstV-5 strains were detected.Furthermore,the two subtypes of HAstV-1 possessed high sequence similarities with a Shanghai strain (FJ375759) and a Russia strain (JF929185),respectively.Conclusions It's indicated that the astrovirus is a major pathogen causing viral diarrhea in Xuzhou area.HAstY-1 is the main genotype,while other genotypes also exist.
