聚酰胺纯化八角残渣黄酮的工艺研究 被引量:9

摘要 目的:研究聚酰胺树脂纯化八角残渣黄酮的工艺。方法:以聚酰胺树脂对八角残渣黄酮的吸附量及解析率为指标,通过静态解吸附实验,确定适于纯化八角残渣黄酮的聚酰胺树脂目数;采用单因素与正交实验优化吸附条件,动态解析实验优化解析条件。结果:聚酰胺纯化八角残渣黄酮最优工艺为上样液浓度为0.05g/m L(生药量),p H为5,聚酰胺柱床高度与内径比为12∶1,流速为1~2BV/h,饱和吸附体积为7.5~8BV,待吸附饱和后,用蒸馏水冲洗聚酰胺树脂柱至Molish反应呈阴性,用4BV体积的90%乙醇洗脱,解析率为70.77%,收集流份液,在60℃下减压回收乙醇至无醇味,干物质中黄酮纯度为87.5%。结论:聚酰胺树脂能有效的纯化八角渣黄酮,最终所得黄酮纯度高,适于工业化生产。 Objective:The aim of this research was to study polyamide purification technology of flavonoids from star aniseed residue. Methods: With the absorption and desorption capacity as indexes,the optimal mesh of polyamide purification for flavonoids was determined from star aniseed residue through static absorption and desorption experiment,the adsorption conditions were optimized from single factor and orthogonal design experiment,the desorption conditions were optimized from dynamic desorption experiments. Results:The optimal purification technology conditions :sample concentration was 0.05g/m L(Crude drug concentration),p H5.0,height-diameter ratio of column 12∶1,flow rate 1~2BV/h,saturated adsorption volume 7.5~8BV,after being saturated adsorption,the polyamide resin column was washed with distilled water until Molish were negative,with 4BV 90% ethanol elution volume,at desorption rate of 70.77%,collect flow of fluid,recycling ethanol at60℃,the purity of flavonoids from dry matter medium was 87.5%. Conclusion:Purified effect of flavonoids from star aniseed residue by polyamide resin was effectively and the finally purity of the flavonoids obtained was high,it was suitable for industrial production.
