摘要
以兰坪虫草无性型(兰坪被毛孢)固体发酵产物为材料,以水为提取溶剂,在常温条件下利用响应面分析法优化虫草多糖和D-甘露醇的超声提取综合工艺。在超声常温水提的单因素实验的基础上,运用响应面法进一步考察液固比、超声时间、超声功率对虫草多糖和D-甘露醇综合提取率的影响,以优化提取工艺。实验得到较优综合提取工艺条件为:液固比43∶1m L/g,超声时间为50min,超声功率为56W。在该工艺条件下虫草多糖提取率为23.06%,D-甘露醇提取率为2.30%。
The ultrasonic extraction technology of polysaccharides and D-mannitol was optimized by response surface methodology at room temperature. The polysaccharides and D-mannitol were extracted by water from the solid fermentation culture of Ophiocordyceps lanpingensis anamorph(Hirsutella lanpingensis). On the basis of single factor ultrasonic extraction in water at room temperature,the effects of ultrasonic time,ratio of liquid to material and ultrasonic power were investigated to extract polysaccharides and D-mannitol,and the technology was optimized by response surface methodology. The results showed that the optimum technology was as follows:the liquid-solid ratio 43∶1m L/g,extraction time 50 min,and extraction power 56 W. Under the conditions,the extraction rates of polysaccharides and D-mannitol were 23.06% and 2.30%,respectively.
