摘要
目的采用柱切换HPLC法测定血浆中的枸橼酸。方法预处理柱为Ultimate CN,分析柱为YMC C18,流动相为0.05mol·L^-1KH2PO4(磷酸调p H2.85),流速1 m L·min-1,检测波长210 nm。血浆样品经过酸化,超滤后进样。结果枸橼酸的线性范围为62.5-1250μg·m L^-1,方法回收率为97.41%-102.52%,批内RSD为3.52%-5.21%,批间RSD为3.07%-4.55%。结论所用方法操作简单,结果准确,重复性好,可用于血浆中枸橼酸的测定。
OBJECTIVE To develop an HPLC method for determination of citric acid in human plasma. METHODS The HPLC system consisted of pretreatment column Ultimate CN and analytical column YMC C1s-The mobile phase was 0.05 mol ·L^-1 KH2PO4 (pH 2.85) with a flow rate of 1 mL.min -1. The detection wavelength was set at 210 nm. The samples were ultrafiltrated after acidified. RESULTS The calibration curves had good linearity within the concentration range of 62.5 - 1250 μg· mL^-1. The average method recoveries for citric acid plasma concentrations were 97.41% - 102.52%. The intra - day RSD and inter - day RSD were 3.52% - 5.21% and 3.07% -4.55%, respectively. CONCLUSION This method provides a sensitive, accurate, precise and reliable analyti- cal procedure for clinical monitoring of citric acid in human plasma.
