摘要
目的采用HPLC法测定茵栀黄口服液中黄芩苷的含量。方法利用中空纤维超滤膜除去样品溶液中的大分子物质;采用Sapphire C18色谱柱,流动相为甲醇-水-磷酸(47∶53∶0.2),检测波长为280 nm,流速1.0mL·min^-1,进样量20μL。结果黄芩苷12.5-200μg·m L^-1与峰面积的线性关系良好(r=0.9998),回收率≥97.2%,RSD≤0.6%。结论所用前处理方法操作简便,可减少大分子物质对色谱柱的损坏。
OBJECTIVE To establish an HPLC method for the determination of baicalin in Yinzhihuang oral liquid. METHODS A hollow fiber based centrifugal ultrafiltration pretreatment procedure combined with HPLC was applied to purify solution samples. And under the effect of the centrifugal force, macromolecules were removed from solution samples. A Sapphire C18 column was used. The mobile phase was methanol - water - phosphoric acid(47 : 53 : 0.2 with a flow rate of 1.0 mL· min^- 1. The detector wavelength was 280 nm, and the injection sample was 20 ~L. RESULTS A good linear relation for baicalin was obtained in the range of 12.5 - 200 p.g.mL^-1(r = 0. 9998). The average recovery of baicalin was over 97.2% with the RSD less than 0.6%. CONCLUSION A hollow fiber based centrifugal ultrafiltration pretreatment procedure is simple,and can reduce the damage of the column.
