摘要
目的探讨聚腺苷酸二磷酸核糖转移酶-1(PARP-1)Val762Ala(T2444C)基因多态性与中国西部甘肃省人群胰腺癌易感性的关系,筛查预测胰腺癌的分子标记。方法选取85例良性胰腺疾病患者(对照组)与66例胰腺癌患者(胰腺癌组),用基质辅助激光解吸电离飞行时间(MALDI-TOF)质谱技术对SNP基因位点进行基因分型,采用χ2-test比较分类变量和各SNP基因分型在病例组和对照组中的分布差异,评价各基因型与胰腺癌发病风险的关系。结果PARP-1 2444C等位基因型(TC+CC)胰腺癌组明显高于良性对照组,TC+CC基因型携带者患胆管癌的风险是TT基因型者的3.4倍(OR 3.377,95%CI:1.715-6.646,P〈0.001)。结论本组数据显示PARP-1 Val762Ala(T2444C)单核苷酸多态性与胆管癌易感性相关,C等位基因型携带者患胰腺癌风险相对较高,PARP-1 Val762Ala(T2444C)单核苷酸多态性可作为预测胰腺癌的生物标志物。
Objective To investigate the association between the PARP-1 Va1762Ala (T2444C) polymorphism and susceptibility to pancreatic cancer in Chinese Han population in area of Gansu Province and explore molecular markers to predict pancreatic cancer. Methods The PARP-1 Va1762Ala (T2444C) polymorphism was analyzed in 66 cases with pancreatic cancer and 85 controls with benign pancreatic diseases by using the matrix-assisted laser desorption/ionization- time of flight mass spectrometry (MALDI-TOF) method. The genotypes of each SNP site were compared between cases and controls by the chi-square test, to evaluate the association between PARP-1 2444 T/C and susceptibility to pancreatic cancer. Results PARP-1 2444 TC genotypes were overrepresented in pancreatic cancer patients compared with controls (OR 2. 476, 95%CI: 1. 159-5. 292, P=0. 018). PARP-1 2444 CC genotype were overrepresented in pancreatic cancer patients compared with controls(OR 5. 778, 95%CI: 2. 278-14. 657, P〈0. 001). Subjects carrying PARP-1 2444 CT/ CC genotypes had a 3.4 fold increased risk for pancreatic cancer compared with TT genotype carriers (OR 3. 377, 95 % CI: 1. 715-6. 646, P〈0. 001). Conclusion PARP-1 Va1762Ala (T2444C) polymorphism is related to higher risk of pancreatic cancer, which increases the susceptibility to pancreatic cancer.
