鱼源嗜水气单胞菌疫苗生产菌株与分离株的抗原性分析 被引量:2

摘要 为分析商品化嗜水气单胞菌(Ah)败血症灭活疫苗生产菌株J-1与近年来分离的YYK090901菌株的抗原性是否存在差异,本研究应用western blot检测了抗J-1血清和抗YYK090901血清识别28株嗜水气单胞菌全菌蛋白情况,并通过细菌凝集试验测定了这2种抗血清与28株菌株的凝集效价。结果表明2种抗血清均能够识别25株菌株的大多数全菌蛋白,但识别GYK1、G060718-2L、Ah120606L 3株菌株的蛋白条带较少,2种抗血清识别同一株菌的全菌蛋白条带不完全一致。抗YYK090901血清对YYK090901、Ah120606L、JX120701具有较高的凝集价(211),对J-1、GYK1等16株菌的凝集价为26~210,其中对J-1、GYK1的凝集价分别为27、210;对N-1-2、TTF20130613L等9株菌的凝集价较低,为24~25。抗J-1血清对1990s年和2012年~2013年分离的大部分菌株均具有较高的凝集价(211~212),仅对GYK1、TTF20130613L等6菌株凝集价较低,为24~25。本研究结果表明,J-1与YYK090901的抗原性存在一定的差异。 Aeromonas hydrophila J-1 strain was a vaccine strain which be used in manufacturing licensed fishery vaccine. To analyse antigenicity difference of A.hydrophila YYK090901 isolate and J-1 strain, Agglutinating test and western blot were used in testing the antibody titers and immunogenicity of the whole bacteria proteins. The western blot profiles revealed more bacteria protein bands from 25 isolates produced strongly specific reaction with anti-J-1 serum and anti-YYK090901 serum, but fewer protein bands were absent in 3 isolates (GYK1, G060718-2L, Ah120606L). The western blot profile of the same isolate with anti-YYK090901 serum was clearly different from that with anti-J-1 serum. The agglutinating antibody titers of anti-YYK090901 serum were 2H to strains YYK090901, Ah1206062, JX120701, respectively, but around 26 to 210 to another 16 strains, including J-l, GYK1 and about 24 to 25 to the other 9 strains. While, the titers of anti-J-1 serum were 211 to 212 to these isolates which were isolated in 1990s and 2012-2013, but only 24 to 25 to another 6 strains. These results demonstrated that the antigenicity of A.hydrophila isolate YYK090901 was different from that of J-1.
