硝菔通结方对便秘大鼠血清5-HT和结肠组织AQP_3的影响 被引量:6

Effects of Xiaofu Tongjie Prescription on the 5-HT content in Serum and the Expression of AQP_3 in Colon in Constipated Rats

摘要 目的:观察硝菔通结方对便秘模型大鼠血清5-HT和结肠组织AQP3表达的影响,揭示该方治疗便秘的作用机制。方法:将50只雄性SD大鼠随机分为正常组、模型组、硝菔通结方低、中、高剂量组。除正常组外,其余各组大鼠以15mg/kg复方地芬诺酯混悬液灌胃28d,复制便秘模型。模型成功后,分别给药21d,后取血清和结肠组织,ELISA法检测血清5-HT含量,Q-PCR法检测结肠组织AQP3的mRNA表达。结果:模型组的大鼠血清中5-HT含量与正常组比较明显升高(P<0.05);药物治疗后,各治疗组血清中5-HT含量均降低(P<0.05),其中硝菔通结方高剂量组最为显著(P<0.01);模型组结肠组织中AQP3与正常组比较表达升高(P<0.05),各剂量的硝菔通结方治疗组均降低了结肠组织中AQP3的表达,其中硝菔通结方高剂量组最为明显(P<0.05)。结论:硝菔通结方可通过降低血清5-HT含量和降低结肠组织AQP3的表达从而促进肠道动力和调节肠道水液代谢,对功能性便秘发挥治疗作用。 Objective:To observe the effect of Xiaofu Tongjie prescription effect on the 5-HT content in serum and the mRNA expression of AQP3 in colon in constipated rats,so as to reveal the mechanism of the prescription in the treatment of constipation.Methods:50male SD rats were randomly divided into normal control group,model group,Xiaofu Tongjie-low dose group,Xiaofu Tongjie-medium dose group and Xiaofutongjie-high dose group.Except the normal control group,the rest of the rats were intragastrically administered with 15mg/kg compound diphenoxylate for 28 days to replicate the constipation model.Then serum and colonic tissue samples were collected after 21 days of treatment.The5-HT content in serum were detected with ELISA method and the mRNA expression of AQP3 were detected with Q-PCR method.Results:The 5-HT content in serum of the model group increased significantly compared with the normal control group(P〈0.05).After prescription treatment,the 5-HT level in each treatment group decreased(P〈0.05),and the Xiaofu Tongjie-high dose group marked decreases(P〈0.01).AQP3 expression in colonic tissue of model group increases compared with the common control group(P〈0.05),different dose of the Xiaofutongjie prescription groups all decrease the AQP3 expression in colonic tissue,but Xiaofu Tongjie-high dose group marked ecreases P〈0.05).Conclusion:Xiaofutongjie prescription can reduce the 5-HT content in serum and decrease the mRNA expression of AQP3 in colon,thus to improve the intestinal power and regulate the water metabolism of intestinal tract.Consequently,it has a great therapeutic effect on functional constipation.
