活血通督汤对脊髓缺血再灌注损伤NF-κB、VCAM-1表达的作用 被引量:15

Effects of Huoxue Tongdu Decoction on inflammatory factor NF-κB and VCAM-1 during spinal cord ischemic-reperfusion injury

摘要 目的:观察活血通督汤对脊髓缺血再灌注损伤关键因子核因子-κB(NF-κB)、血管细胞黏附分子-1(VCAM-1)表达的影响,探讨活血通督汤对脊髓缺血再灌注损伤作用机制。方法:将新西兰家兔随机分为假手术组(8只)、模型组(32只)和活血通督汤组(32只)。3组术前7d均予连续灌胃,活血通督汤组灌服活血通督汤,每日2次,假手术组及模型组予灌服0.9%氯化钠溶液,每日2次。假手术组分离出L3-6动脉但不予夹闭,其余2组分离并夹闭L3-6动脉,制成脊髓缺血再灌注损伤模型。3组分别于术后0.5、1、4、8h取出家兔脊髓,采用免疫组化法和ELISA法检测脊髓组织中的NF-κB、VCAM-1的含量。结果:NF-κB在同一时点表达,假手术组最少,模型组最高;与假手术组相比,其余2组各时点NF-κB都明显升高(P<0.01);但活血通督汤组的表达水平显著低于模型组(P<0.05,P<0.01);VCAM-1在同一时点,假手术组表达最少,模型组表达最高;与假手术组相比,其余2组各时点VCAM-1的表达显著升高(P<0.01);血管表达率情况观察:假手术组始终无阳性血管表达,0.5h时3组皆无阳性血管表达,1、4、8h模型组阳性血管表达率明显高于活血通督汤组(P<0.05,P<0.01)。结论:活血通督汤能有效改抑制NF-κB、VCAM-1的表达,从而减轻炎性反应,以期达到减轻脊髓缺血再灌注损伤。 Objective: To observe the effects of Huoxue Tongdu Decoction on inflammatory factor NF-κB and VCAM-1 during spinal cord ischemic-reperfusion injury and explore its mechanism.Methods: New Zealand rabbits were randomly divided into three groups: sham-operation group(n=8),normal saline group(n=32) and Huoxue Tongdu Decoction group(n=32).For 7 days of pre-operation,Huoxue Tongdu Decoction group was gavaged with Huoxue Tongdu Decoction twice a day,while sham-operation group and normal saline group were both gavaged with normal saline twice a day.The sham-operation group was separated the L3-6 artery but not clipping,while other groups were separated the L3-6 artery to establish the ischemia reperfusion injury model of spinal cord.At 0.5,1,4,8 hours of post-operation,spinal cord of all rabbits were extracted.The immunohistochemistry method and ELISA were used to detect the contents of NF-κB and VCAM-1 in spinal cord.Results: The expression of NF-κB in sham-operation group was the lowest,normal saline group was the highest.Compared with the shamoperation group,the expressions of NF-κB in the other groups were obviously increased at different time(P〈0.01).NF-κB in Huoxue Tongdu Decoction group was significantly lower than normal saline group(P〈0.05,P〈0.01).At the same point,the expression of VCAM-1 in sham-operation group was the lowest and normal saline group was the highest.Compared with the sham-operation group,the expression of VCAM-1 in the other groups were obviously increased at different time(P〈0.01).The observation in vascular expression rate: there was no positive vascular expression in sham-operation group and in three groups at 0.5 hour.But,positive vascular expression in normal saline group was significantly more than Huoxue Tongdu Decoction group at 1st,4h and 8h(P〈0.01).Conclusion: Huoxue Tongdu Decoction could inhibit the expressions of NF-κB and VCAM-1 to alleviate the inflammatory reaction,which would help to reduce the ischemic-reperfusion injury of spinal cord.
