消痈溃得康抑制局部炎性反应促进胃溃疡愈合的研究

Effects of the Xiaoyong Kuidekang on inhibiting local inflammation and promoting healing of gastric ulcer

目的:探讨消痈溃得康对乙酸诱导大鼠胃溃疡模型治疗作用及其机制。方法:将大鼠随机分为正常组、假手术组、模型组、溃得康组和奥美拉唑组。采取胃黏膜局部投予乙酸方法制备胃溃疡模型,溃得康组、奥美拉唑组术后第2天开始给药,假手术组、模型组给纯水。术后第12天取材测量胃溃疡面积,取胃组织进行病理切片,HE染色,观察黏膜炎性反应情况;酶联免疫法检测胃组织匀浆白介素(IL)-6、IL-8及肿瘤坏死因子-α(TNF-α)的含量。结果:假手术组与正常组均无溃疡形成,病理未见炎性细胞浸润。模型组大鼠胃黏膜局部溃疡,溃疡底部附苔,溃疡周边红肿;溃疡部位组织切片病理染色可见黏膜上皮脱落,黏膜下层大量的炎性细胞浸润;溃得康组、奥美拉唑组与模型组比较,溃疡面积明显减小(P<0.05),黏膜下层的炎性细胞浸润明显减轻(P<0.01)。模型组与假手术组比较,胃组织匀浆IL-6、IL-8及TNF-α含量明显升高(P<0.01);溃得康组、奥美拉唑组与模型组比较,胃组织IL-6、IL-8及TNF-α含量明显降低(P<0.01)。结论:消痈溃得康促进乙酸诱导大鼠胃溃疡模型溃疡愈合,减轻局部炎性细胞浸润,降低胃组织匀浆IL-6、IL-8及TNF-α的水平。

Objective： To study the effect of Xiaoyong Kuidekang on the treatment of rat gastric ulcer model and its molecular mechanism.Methods： Rats were randomly divided into 5 groups： normal group,sham group,model group,Kuidekang group and omeprazole group.The gastric ulcer model was established with acetic acid injuring local gastric mucosa.After 2 days of operation,Kuidekang group and omeprazole group were given corresponding drugs,while the sham group and model group were given pure water.After 12 days of operation,the rats were sacrificed.The area of gastric ulcer was measured.HE staining was used to observe the inflammation respond in gastric mucosa.ELASE was used to detect the contents of IL-6,IL-8 and TNF-αin stomach tissue.Results： There was no gastric ulcer and inflammatory cell infiltration in the normal group nor in the sham group.There was local ulcer in gastric mucosa in the model group.The ulcer attached music At the bottom,and in the surrounding,the ulcer was red and swollen.HE staining showed that mucous epithelium fell off and submucosa was infiltrated with vast of inflammatory cells.Compared with the model group,the areas of gastric ulcer in Kuidekang group and omeprazole group was decreased（P〈0.05）,and inflammatory cell infiltration in submucosa as well（P〈0.01）.The results of contents of IL-6,IL-8 and TNF-α in stomach tissue showed that there was no difference in the contents of IL-6,IL-8 and TNF-α between sham group and normal group.The contents of IL-6,IL-8 and TNF-α in model group were obviously higher than those in sham group（P〈0.01）.The contents of IL-6,IL-8 and TNF-α in Kuidekang group and omeprazole group were lower than those in model group（P〈0.01）.Conclusion： Xiaoyong kuidekan could promote the healing of gastric ulcer induced by acetic acid,alleviate the local inflammatory cell infiltration and decrease the levels of IL-6,IL-8 and TNF-α in stomach tissue.
