摘要
目的:探讨小鼠在脾虚状态下对消化道感染白念珠菌的易感性及其免疫学机制,为防治白念珠菌感染和脾虚证提供一定的实验依据。方法:采用饮食失节加劳倦过度的复合因素制备小鼠脾虚证模型,经消化道感染白念珠菌,检测粪便中的活白念珠菌数,应用RT-PCR方法检测肠组织局部V-干扰素(IFN-γ)mRNA和白细胞介素-10(IL-10)mRNA的表达水平。结果:与正常感染组比较,自第6天起脾虚感染组小鼠粪便活菌数持续显著增高(P<0.05,P<0.01)。与脾虚模型组比较,正常感染组与脾虚感染组小鼠在感染后期肠组织表达IFN-γ mRNA和IL-10 mRNA的水平均显著升高(P<0.01);与正常感染组比较,脾虚感染组小鼠在感染后期IFN-γ mRNA的表达水平显著升高(P<0.01),但IL-10 mRNA表达显著减少(P<0.05)。结论:消化道的白念珠菌感染是否致病与小鼠的免疫力密切相关,尤其是在免疫功能低下时危害更大,同时又进一步加重脾虚状态或感染的病情。
Objective: Investigation of the susceptibility of Candida albicans on alimentary tract in the state of deficiency of spleen to explore the immunological mechanisms,and to provide an experiment reference for the prevention and treatment of Candida albicans infection and deficiency of spleen syndrome.Methods: The animal model of spleen deficiency syndrome was established by irregular diet and overstrain methods,and the mice were infected by Candida albicans through alimentary tract.The number of living Candida albicans in faeces was detected.RT-PCR method was used to detect the mRNA expression of IFN-γ and IL-10 in the intestinal tissue.Results: The number of living Candida albicans in faeces were increased in spleen deficiency infection group.The mRNA expression of IFN-γ and IL-10 in normal infection group and spleen deficiency infection group were both significantly higher than those in spleen model group at the late stage of infection(P〈0.01).The mRNA expression of IFN-γ in spleen deficiency infection group was both lower than that in normal infection group at the late stage of infection(P〈0.05).Conclusion: The Candida albicans infection is closely related to the immune system of mice.When the immune system is weakened,the Candida albicans infection would be more harmful to the body,which further aggravated the deficiency state and infection.
