半夏泻心汤及其拆方诱导胃腺癌SGC-7901细胞凋亡机制 被引量:10

Research on mechanism of Banxia Xiexin Decoction and its components induced-apoptosis in human gastric adenocarcinoma SGC-7901 cell

摘要 目的:研究半夏泻心汤及其拆方诱导胃腺癌SGC-7901凋亡机制。方法:把半夏泻心汤拆分为苦降、辛开、补益3个组方,采用S-P免疫组化法检测半夏泻心汤及其拆方凋亡相关基因STAT3、P-STAT3、Bax、Bcl-2的表达。结果:高浓度的半夏泻心汤及其拆方作用于SGC-7901胃癌细胞48h后均能使Bax的蛋白表达增强,使其Bcl-2蛋白的表达降低,低浓度效果不明显,补益组则无效。结论:半夏泻心汤全方及苦降、辛开组方诱导细胞凋亡机制可能与通过干预STAT3转录因子增高Bax的表达,降低Bcl-2的表达有关。 Objective: To observe the mechanism of Banxia Xiexin Decoction and decomposed recipes effect on SGC-7901 tumor cell apoptosis.Methods: Banxia Xiexin Decoction was divided into Pungent openning(dried ginger,pinellia ternate) group,Bitter Descending(coptis chinensis,scutellaria) group and Invigoration(glycyrrhiza,ginseng) group by its different nature and flavor.The S-P(Phosphate-Peroxidase) immunohistochemical method to detect the expression of Apoptosis Associated Gene STAT3,P-STAT3,Bax,bcl-2 of Banxia Xiexin Decoction and its decomposed recipes in SGC-7901 tumor cell line.Results: Both Banxia Xiexin Decoction and its decomposed recipes can induce the increase of the Bax Protein expression and the decrease of the Bcl-2 protein expression.While invigoration group had no such effect.Conclusion: Banxia Xiexin Decoction and its compositions Bitter Descending group and Pungent opening group can induce cell apoptosis which may related to interfering STAT3 transcription factor to increase Bax expression and reduce Bcl-2 expression.
