摘要
目的:初步探讨桑叶有效部位对胰岛素抵抗(IR)HepG2细胞模型的保护作用及机制。方法:测定HepG2细胞的生长曲线,成功复制HepG2细胞IR模型后,采用各降糖有效部位的提取液进行干预,并与常用的罗格列酮和参芪降糖胶囊进行对照,测定各组药液对HepG2细胞及IRHepG2细胞增殖的影响,观察比较各给药组细胞的葡萄糖消耗量。结果:筛选出2.5-10μg/L的罗格列酮、5-100mg/L的参芪降糖胶囊、2-12mg/L的桑叶生物碱、5-100mg/L的桑叶黄酮、5-100mg/L的桑叶多糖、5-100mg/L的桑叶水提物对HepG2细胞及IRHepG2细胞增殖无影响,每组给予不同浓度的药物干预后,高、中、低剂量的生物碱、水提物组和高剂量的黄酮、多糖组可显著增加其葡萄糖消耗量(P<0.01)。结论:桑叶生物碱具有显著的改善模型细胞IR的作用,可能与其增加HepG2细胞的葡萄糖消耗量和促进葡萄糖的吸收及摄取有关。
Objective: To investigate the protective effect and its mechanism of different effective extracts of mulberry leaves on the insulin resistance(IR) model of HepG2 cells.Methods: After the growth curve of HepG2 cells was measured and the IR model was duplicated successfully,the IR model was intervened with the solution of different effective extracts of mulberry leaves,and compared with Rosiglitazone and Shenqi Jiangtang capsules,to determine the influence on the cell proliferation of HepG2 cells and IR HepG2 cells,to study the effect of glucose consumption in each group.Results: Each treatment group had no effect on the cell proliferation of HepG2 cells and IR HepG2 cells.After intervened with different concentrations of the drug,the alkaloids(2-12mg/L),aqueous extract group(5-100mg/L) and high dose of flavonoids(5-100mg/L) and polysaccharides group(5-100mg/L) could increase the glucose consumption(P〈0.01).Conclusion: The alkaloids of mulberry leaves had the appreciable effect on improving the insulin resistance of IR HepG2 cell model.Its mechanism is probable related with increasing the IR HepG2 cell model's glucose consumption and promoting glucose absorption and ingestion.
