补骨脂干预兔激素性股骨头坏死空骨陷窝的影响 被引量:14

Effects of Fructus Psoraleae on empty bone lacuna of osteonecrosis of the femeral head induced by hormone in rabbit

摘要 目的:以马血清加甲强龙造模激素性股骨头坏死模型为研究对象,研究补骨脂治疗激素性股骨头坏死的初步效果和作用机制,为其临床应用提供理论依据。方法:选用日本大耳白兔30只,马血清+甲基强的松龙建立激素性股骨头坏死兔模型,分为空白组、模型组、治疗组3组进行比较。造模完成后,空白组,模型组每天用0.9%氯化钠溶液10mL灌胃1次,治疗组用补骨脂提取液10mL灌胃1次。8周后,解剖出股骨头并观察股骨头形态,色泽,脱钙,包埋,切片,HE染色,光镜下检测各组骨陷窝数,计算空骨陷窝比率,并进行不同组间比较。结果:补骨脂可促进成骨细胞增殖,抑制破骨细胞分离,光镜下可见空白组和治疗组空缺骨陷窝率显著低于模型组(P<0.05),股骨头骨组织再生以达到防治骨质疏松的作用,进一步达到治疗股骨头坏死,延缓塌陷的目的。结论:补骨脂提取液能够促进体外成骨细胞增殖及分化成熟,抑制破骨细胞活性、促进股骨头骨再生,改变股骨头内环境向好的方向转变,防治激素引起的骨质疏松,为其能够治疗激素引起的股骨头坏死提供了初步理论依据。 Objective: To study the effects and mechanism of treating osteonecrosis of the femeral head induced by hormone with Fructus Psoraleae based on the rabbit model of osteonecrosis of the femeral head induced by horse serum combined with methylprednisolone,and to provide theoretical foundation for clinical application.Methods: Thirty Japanese white rabbits(1-1.5kg) were provided by animal experiment center of Henan province.The rabbits were divided into a control group,a model group and a treatment group.Rabbits in the model group and the treatment group were intervened with horse serum combined with methylprednisolone to establish the animal model of osteonecrosis of the femeral head.After animal model established,rabbits in the control group and the model group received gavage administration with normal saline(10m L) daily,and rabbits in the treatment group with extracting solution of Fructus Psoraleae(10mL).Eight weeks later,the form,color and luster of femoral head of rabbits were observed.Femoral heads were decalcified,embedded and cut into slices.Femoral head sections were stained with HE coloration.Empty lacunae number was observed under light microscope,and the empty lacunae ratio was calculated and compared among different groups.Results: Fructus Psoraleae could promote osteoblast proliferation and inhibit osteoclast separation.The empty lacunae ratio of the control group and the treatment group was significant lower than that of the model group(P〈0.05).Bone regeneration of femoral head could prevent and treat osteoporosis so as to treat osteonecrosis of femoral head further and delay the collapse.Conclusion: Extracting solution of Fructus Psoraleae could promote proliferation,differentiation and maturity of osteoblast,and it also could inhibit osteoclast activity,promote femoral head bone regeneration,and improve femoral head internal environment so as to prevent and treat osteoporosis induced by hormone and provide preliminary theoretical basis for the treatment of osteoporosis induced by hormone.
