摘要
目的探讨人脐带间充质干细胞(h UCMSCs)裂解液对食管癌EC9706细胞中尿激酶型纤溶酶原激活物(u PA)及基质金属蛋白酶-2(MMP-2)表达的影响。方法收集h UCMSCs,采用反复冻融法制备h UCMSCs裂解液;不同浓度h UCMSCs裂解液作用于食管癌EC9706细胞60 h,对照组加入体积分数10%胎牛血清的培养液。采用Western blot法检测食管癌EC9706细胞中u PA、MMP-2的表达。结果加入h UCMSCs数<食管癌细胞数的裂解液时,食管癌EC9706细胞中u PA、MMP-2的表达与对照组比较差异无统计学意义(P>0.05);加入h UCMSCs数≥食管癌细胞数的裂解液时,食管癌EC9706细胞中u PA、MMP-2的表达较对照组显著降低(P<0.05)。结论 h UCMSCs裂解液对食管癌EC9706细胞的迁移抑制作用可能与u PA、MMP-2有关。
Objective To investigate the effect of the lysate from human umbilical cord mesenchymal stem cells( h UCMSCs) on the expression of urokinase plasminogen activator( u PA) and matrix metalloproteinases-2( MMP-2) in esophageal carcinoma EC9706 cells. Methods h UCMSCs were collected and cell lysate was prepared through repeated freeze-thaw cycles. EC9706 cells were treated by cell lysates with different cell numbers of h UCMSCs for 60 hours while the control group was treated with the 10% fetal bovine serum. The expression of u PA and MMP-2 in EC9706 was detected by Western blot. Results The lysate from h UCMSCs with numbers less than treated EC9706 cells did not change both u PA level and MMP-2 level in EC9706 cells when compared with control group( P 0. 05). The expression of u PA and MMP-2 in EC9706 cells was decreased significantly compared with control group when the lysate from h UCMSCs with the number of cell was equal or greater than treated EC9706 cells( P 0. 05). Conclusion h UCMSCs lysate can inhibit the migration of esophageal carcinoma EC9706 cells,which may be associated with u PA and MMP-2.
