红花多糖对人胃癌细胞SGC-7901凋亡的形态学实验研究 被引量:12

摘要 目的:从形态学角度探讨红花多糖(SPS)诱导人胃癌细胞SGC-7901凋亡的作用。方法:MTT法观察SPS对人胃癌SGC-7901细胞体外增殖的抑制作用。荧光显微镜和透射电镜观察细胞凋亡的形态学变化。Rhodamine 123染色观察SPS处理后细胞线粒体跨膜电位变化。结果:SPS能明显抑制SGC-7901细胞增殖,具有时间和剂量依赖性。显微镜下发现SPS处理后贴壁细胞减少,部分细胞变圆、皱缩、脱落,部分细胞崩解。荧光显微镜下和透射电镜下,SPS作用的细胞呈现典型凋亡细胞状态。胃癌细胞Rhodamine 123荧光强度明显减弱。结论:SPS对人胃癌细胞体外增殖有抑制作用,且具有明显的时间和剂量依赖性。 Objective:From the view of morphology to study the effect of Safflower polysaccharide (SPS) on the apoptosis of gastric carcinoma cell line SGC -7901. Methods:Inhibition of SPS on the proliferation of human gastric carcinoma cell line SGC -7901 was determined by methyl thiazolyl tetrazolium chromatometry. Mor- phology of cells was observed with fluorescent microscope and transmission electron microscope. Changes of mi- tochondria membrane potential of the cells treated by SPS were detected by Rhodamine 123 staining. Results: SPS could obviously inhibit gastric carcinoma eel1 line SGC -7901 to proliferate, and this inhibition relied on time and dosage conspicuously. SGC7901 treated by SPS showed less adherent cells, and some cells became round, shrank, shed from culture flake wall and some ceils were disrupted. SGC7901 cells presented typical apoptposis under fluorescence microscope and transmission electron microscope. Fluorescence intensity of Rho- damine 123 was significantly decreased in SGC -7901 cells. Conclusion:SPS can inhibit human gastric carci- noma cell to proliferate and this inhibition has time and dose dependence.
