摘要
目的:探讨经方薯蓣丸对变应性鼻炎(AR)大鼠鼻黏膜AQP5表达的影响。方法:70只大鼠随机分为正常对照组、模型组、通窍鼻炎片组、辛芩颗粒组、经方薯蓣丸高、中、低剂量组,每组10只。采用OVA复制AR大鼠模型。建模成功后,治疗组分别采用不同浓度薯蓣丸浓缩水煎液(3.2、1.6、0.8g/0.1kg)灌胃。治疗14天后,比较治疗前后各组大鼠行为学积分变化情况,检测各组大鼠鼻黏膜AQP5蛋白的表达情况。结果:造模后各组大鼠行为学积分均高于正常对照组(P<0.01),治疗后各组行为学积分较模型组均有降低(P<0.01);免疫组化染色显示模型组平均光密度值(MOD)低于正常对照组(P<0.01);除辛芩颗粒组外,各治疗组MOD值较模型组均有升高(P<0.01);高、中剂量组MOD值高于低剂量组及通窍鼻炎片组(P<0.01),而高、中剂量组间比较,无统计学意义(P>0.05)。结论:经方薯蓣丸对AR大鼠疗效显著。上调鼻黏膜中AQP5蛋白的表达可能是该方治疗AR良好疗效的作用机理之一。
Objective: To study the effect of classical prescription Shuyuwan on the expression of AQP5 in nasal mucosa of allergic rhinitis rats.Methods: 70 rats were randomly divided into a normal control group,a model group,a Tongqiao-biyan tablet group,a Xinqin granule group,Shuyuwan high,medium and low doses groups,with 10 rats in each group.Use OVA to copy the model of AR rats.After the success of modeling,the treatment groups were treated with different concentrations of Shuyuwan decoction(3.2,1.6,0.8g /0.1kg) by gavage.After 14 d intervention,compare behavioral changes integral before and after treatment,and expression of AQP5 protein in nasal mucosa of rats.Results: The integral behavior of rats after modeling was higher than that in the normal control group(P〈0.01),after the intervention behavior integral was reduced compared with those in the model group(P〈0.01).Immunohistochemical staining showed that in the model group,the average optical density(MOD) was lower than that in the normal control group(P〈0.01); In addition to Xinqin granule group,the treatment groups' MOD value was higher than that in the model group(P〈0.01);high and medium dose groups' MOD values were higher than those in the low dose group and Tongqiao Biyan tablet group(P〈0.01).Relatively high and medium dose groups' MOD values were not statistically significant(P 〉0.05).Conclusion: Shuyuwan has a significantly therapeutic effect on AR rats.Upregulating AQP5 expression in nasal mucosa may be one of the mechanisms for the treatment of AR.
