摘要
目的:对刺五加有效部位进行体外药效学评价。方法:采用MTT法检测细胞存活率,流式细胞仪测细胞凋亡率,紫外可见分光光度计测LDH的漏出量以及SOD、MDA、NO、NOS的活性和含量。结果:刺五加有效部位可以发挥良好的神经元保护作用,能提高MPP+损伤PC12细胞的存活率(P<0.01),降低细胞凋亡率(P<0.01),降低LDH的漏出量以及NO、NOS、MDA的含量和降低SOD活性,有统计学意义(P<0.05)。结论:刺五加有效部位对MPP+损伤PC12细胞具有一定的保护作用,其作用机制可能是通过降低PC12细胞NO、NOS、MDA的含量和LDH的漏出量,引起胞内SOD的活性增强,从而降低细胞凋亡率,提高细胞存活率而实现的。
Objective: To study the pharmacodynamic evaluation of effective part of Acanthopanacis senticosus Harms( AS) using an in vitro model of Parkinson's disease( PD). Methods: The pharmacodynamics studies were undertaked by the indexes of cell viability detected by MTT,apoptosis rate measured by flow cytometry,LDH leakage and the activities and contents of SOD,MDA,NO、NOS detected by UV-visible spectrophotometer. Results: The pharmacodynamics experiment certified that effective part of AS had protective effect on the damage of PC12 cells induced by MPP^+. This was expressed by enhancing the cell survival rate,and its mechanisms maybe implement through the reduction of the leakage of LDH and lowering the contents and inhibiting the activity of NO,NOS,MDA,and all the above were functioned in inhibiting apoptosis in PC12 cells. Conclusion: This study demonstrates that the effective part of AS have neuroprotective activity and provides an evidence-base for the treatment of AS on PD.
