摘要
目的:观察、探讨壮骨止痛方对去势雌鼠骨质疏松模型的疗效。方法:选取40只3月龄SD雌性大鼠,按体质量随机分为4组,10只/组,分别为正常对照组、假手术组、模型组、壮骨止痛方组。术后5d拆线后开始给药,连续l3周后处死。取血和骨标本从骨密度、血清雌二醇(E2)、甲状腺激素(T3、T4)、促甲状腺激素(TSH)和甲状旁腺激素(PTH)等几个方面研究其疗效。结果:模型组与空白组相比,大鼠血清E2、骨小梁密度明显降低,差异显著(P<0.01),表明造模成功。壮骨止痛方组与模型组相比,血清中E2浓度明显增加,T3、T4、TSH浓度降低,PTH浓度明显降低,并能明显增加平均骨小梁密度(P<0.01)。结论:壮骨止痛方能降低血清T3、T4、TSH、PTH中水平的趋势.增加壮骨止痛方组大鼠平均骨小梁密度及血清E2浓度,调整垂体-甲状腺轴功能,从而抑制骨吸收,促进骨形成,降低骨转换率,纠正骨代谢的负平衡能有效预防及治疗骨质疏松症。
Objective: To observe and discuss the effect of Zhuanggu Zhitong Prescription on castrated female osteoporosis model. Methods: Only 3 months aged 40 SD female rats,according to the weight,were randomly divided into four groups,10 in each group,normal control group,control group,model group,Zhuanggu Zhitong Prescription group. Five days after stitches of the operation,different treatments were carried out for 13 weeks. Blood and bone samples were collected for investigations on bone mineral density,estradiol( E2),thyroid hormone( T3,T4),thyroid-stimulating hormone( TSH) and parathyroid hormone( PTH) and other aspects to evaluate efficacy. Results: After castration,compared with the blank group,the rats serum E2 was significantly decreased and had significant statistical significance compared with that of the experimental group( P〈0. 01). The model group's femoral trabecular bone density was significantly lower than that of the control group with significant difference( P〈0. 01),indicating the successful modeling. Compared with model group,Zhuanggu Zhitong Prescription group's serum E2,T3,T4,TSH and PTH concentrations were on the decline and could significantly increase the average trabecular bone density( P〈0. 05). Conclusion: After castration,compared with the blank group,the rats serum E2 was significantly decreased. Compared with the experimental group,there was significant statistical significance( P〈0. 01). The model group's femoral trabecular bone density was significantly lower than that of the control group with significant difference( P〈0. 01),indicating the successful modeling. Zhuanggu Zhitong Prescription can reduce serum E2,T3,T4,TSH and PTH levels and increase the trabecular bone density and adjust the pituitary-thyroid axis function,and thus inhibiting bone reabsorption,promoting bone formation,reducing bone conversion rate and correcting negative balance of bone metabolism. It can effectively prevent and treat osteoporosis.
