联合应用自噬抑制剂增加雷帕霉素引起的肺癌细胞凋亡 被引量:1

摘要 目的探讨自噬抑制剂对雷帕霉素引起的肺癌细胞凋亡的影响。方法选用人肺癌A549细胞,MTT方法检测单独利用雷帕霉素(Rapamycin)或者联合自噬抑制剂3-MA对A549细胞存活率的影响,Western印迹检测单独利用雷帕霉素(Rapamycin)或者联合自噬抑制剂3-MA凋亡相关蛋白Caspase-3和PARP,以及线粒体相关蛋白细胞色素C表达水平的影响。结果 MTT结果显示,Rapamycin可以明显地引起A549细胞存活率的下降,并呈现出剂量依赖性。Western印迹结果显示,Rapamycin可以引起凋亡相关蛋白酶切的PARP、酶切的Caspase-3以及线粒体凋亡相关蛋白细胞色素C(cytochrome c)表达水平明显的上升。MTT结果显示,联合应用自噬抑制可以明显的增加Rapamycin引起的细胞存活率的下降。Western印迹显示,与单独给予Rapamycin相比较,联合应用自噬抑制剂3-MA,凋亡相关蛋白酶切的PARP、酶切的Caspase-3进一步增加。结论自噬抑制剂3-MA可提高人肺癌A549细胞对m TOR抑制剂Rapamycin的敏感性。
