摘要
目的探讨肾茶水提取物对腺嘌呤致慢性肾功能衰竭(CRF)大鼠的肾脏保护作用及其可能的机制。方法采用腺嘌呤灌胃法建立大鼠CRF模型,同时用肾茶灌胃治疗。治疗第17d测定尿液尿酸含量;4周后,采血测血清肌酐(Cr)、尿素氮(BUN)、丙二醛(MDA)含量,并测定菊粉清除率代表肾小球滤过功能。取大鼠肾脏进行H-E染色病理组织学观察和肾组织免疫组织化学染色检测碱性成纤维细胞生长因子(bFGF)的表达。结果与模型对照组比较,肾茶治疗组肾脏病理改变轻,尿液尿酸含量增加,血清Cr、BUN、MDA含量降低,菊粉清除率升高,bFGF表达减少(均为P<0.05)。结论肾茶对CRF大鼠的肾脏有保护作用,其主要作用机制可能与肾茶增加尿液尿酸排泄、抗氧化作用和抑制肾小管及肾间质bFGF表达有关。
Objective To investigate the renal protective effect and possible mechanism of aqueous extract from a herb C .Spicatus on chronic renal failure rats induced by adenine . Methods The animal model of CRF was established by the adenine gastric infusion method . The rats were treated with ig&amp;nbsp;C .Spicatus daily . The contents of urine uric acid were detected on the 17 d of treatment . BUN ,Cr , MDA of blood serum were respectively detected ,as well as determined GFR after 4 weeks . The kidneys were observed for the histopathology by H‐E dyeing and detected the level of bFGF by immuhistochemis‐try dyeing . ResuIts The pathology changes of the C .Spicatus groups were lighter than those of the model control group . The contents of urine uric acid of C .Spicatus group were significantly higher than those of model control group(P&lt;0 .05) . BUN ,Scr ,MDA were lower than those of model control group (P&lt;0 .05) . The inulin clearance rate of C .Spicatus group were higher than those of model control group (P&lt;0 .05) . The expression of bFGF in C .Spicatus group was significantly less than it in model control group(P&lt;0 .05) . ConcIusion C .Spicatus had protective and promoted function on CRF in rat kidney induced by adenine . For the performance of C .Spicatus could reduce serum Cr ,BUN and increased glo‐merular filtration rate ,reduce the renal pathological changes . T he effective mechanism may be associated with the its accrescence of uric acid evacuation ,improvement of glomerular filtration rate and anti‐oxyda‐tion activity . In addition ,it might relate to inhibitory effect of renal tubule and interstitium of bFGF expression .
