黄芪总黄酮与丹参酮ⅡA磺酸钠对病毒性心肌炎小鼠心肌细胞内质网伴侣蛋白及L-型钙通道表达的作用 被引量:20

摘要 目的:观察黄芪总黄酮(TFA)与丹参酮ⅡA磺酸钠对病毒性心肌炎(VMC)小鼠心肌细胞内质网伴侣蛋白GRP78及L-型钙通道(LCCs)mRNA表达的影响。方法:48只雄性Balb/c小鼠分为对照组、VMC组、TFA组和丹参酮ⅡA磺酸钠治疗组(丹参组),每组12只。注射柯萨奇(CV)B3病毒10d处死小鼠取心脏行心肌病理观察,采用半定量逆转录-聚合酶链式反应(RT-PCR)检测对照组、VMC组、TFA组、丹参组心肌细胞LCCsɑ1亚单位mRNA表达量,采用Western blotting技术检测4组心肌细胞内质网伴侣蛋白GRP78表达量。结果:1与对照组比较,VMC组心室肌LCCsɑ1亚单位的mRNA表达量明显增多(P<0.01);与VMC组比较,TFA可使VMC小鼠心室肌细胞LCCsɑ1亚单位的mRNA表达量减少(P<0.01),而丹参酮ⅡA磺酸钠可使VMC小鼠心室肌细胞LCCsɑ1亚单位的mRNA表达量减少(P<0.01)。2与对照组比较,VMC组心室肌细胞内质网伴侣蛋白GRP78表达量明显增多(P<0.01);与VMC组比较,TFA可使VMC小鼠心室肌细胞内质网伴侣蛋白GRP78表达量减少(P<0.01),而丹参酮ⅡA磺酸钠可使VMC小鼠心室肌细胞内质网伴侣蛋白GRP78表达量减少(P<0.05)。结论:TFA与丹参酮ⅡA磺酸钠可以降低VMC小鼠心室肌细胞LCCsɑ1亚单位的mRNA表达量,缓解VMC小鼠心肌细胞内质网应激,抑制心肌细胞钙超载介导的内质网应激,从而改善心肌损伤。 Objective:To observe the effects of total flavonoids of astragalus(TFA)and sodium TanshinoneⅡ-A sulfonate(TanshinoneⅡ-A)on gene expression of L-type calcium channels(LCC)and expression of endoplasmic reticulum chaperones glucose-regulated protein(GRP)78in viral myocarditis.Method:Forty eight male Balb/c mice were randomly divided into 4groups:control group(n=12),viral infection control group(n=12),TFA group(n=12),TanshinoneⅡ-A group(n=12).When all the mice were sacrificed on day 10,the gene expression of LCCα1subunit was monitored by RT-PCR and the expression change of endoplasmic reticulum GRP78 of cardiac muscle cells was monitored by western blot.Result:Compared with that of virus infection control group,the gene expression of LCCα1subunit of ventricle muscle cell in TFA group was reduced(P0.01),the expression of endoplasmic reticulum chaperones GRP78 was dropped(P0.01).Compared with that of virus infection control group,the gene expression of LCCα1subunit of ventricle muscle cell in Tanshinone Ⅱ-A group was reduced(P0.05),the expression of endoplasmic reticulum chaperones GRP78TanshinoneⅡ-A was dropped(P0.05).Conclusion:TFA and TanshinoneⅡ-A suppress myocardial endoplasmic reticulum stress in viral myocarditis mouse,so they can protect the myocardial damage.
