摘要
目的研究小鼠Bin1b真核表达重组质粒pSG.SS.C3d3.YL.Bin1b的DNA疫苗雄鼠免疫避孕效应。方法应用pSG.SS.C3d3.YL.Bin1b(C3d3-Bin1b组)肌注免疫雄鼠,观察其精子活力及避孕效应,ELISA法检测雄鼠血清抗Bin1b蛋白IgG抗体最大稀释度、IL-2、INF-γ、IL-4、IL-10水平。PBS组、空载体pSG.SS.YL组(空载体组)和pSG.SS.YL.Bin1b组(Bin1b组)为对照组。结果 C3d3-Bin1b组雄鼠精子活力、雌鼠受孕率、平均产仔数量显著降低,免疫避孕效果显著好于Bin1b组。C3d3-Bin1b组雄鼠血清抗Bin1b蛋白IgG抗体滴度显著升高,血清IL-2、INF-γ浓度显著降低,血清IL-4、IL-10浓度显著增加。结论真核表达重组质粒pSG.SS.C3d3.YL.Bin1b可有效诱导雄鼠产生免疫性避孕效应,效果优于pSG.SS.YL.Bin1b。
Objective To explore the immunological contraceptive effects induced by pSG. SS. C3d3. YL. Binlb on male mice. Methods After male mice were immunized with pSG. SS. C3d3. YL. Binlb, their serum IgG titres against Binlb, the content of serum IL-2,INF-7, IL-4 and IL-10 were detected using ELISA assays. PBS group, pSG. SS. YL group and pSG. SS. YL. Binlb group (Binlb group) served as control groups. Results The sperm viability, pregnancy rate of the female mice, and average litter number of the C3d3-Binlb group were significantly lower than those of the Binlb group. In the C3d3-Binlb group,serum IgG titres was higher, the content of IL-2 and INF-7 decreased significantly, while the content of IL-4 and IL-10 in- creased markedly. Conclusion pSG. SS. C3d3. YL. Binlb has better contraceptive effects on male mice than pSG. SS. YL. Binlb.
