摘要
目的探讨烯酯酰辅酶A水解酶1(enoyl coenzyme A hydratase 1,Ech1)表达变化在肝癌中的意义及Ech1的亚细胞定位与肿瘤恶性生物学行为的关系。方法应用免疫组化法检测20例正常肝组织和30例原发性肝癌组织中Ech1的表达;应用亚细胞分离法检测Ech1在Hca-F和FEch1下调细胞的四种不同亚细胞组分中的表达。结果 Ech1在原发性肝癌中的表达高于正常肝组织,差异有统计学意义(P<0.05);Ech1表达与患者年龄、AFP值及是否伴有肝硬化或伴有病毒性肝炎差异无统计学意义(P>0.05)。Ech1在Hca-F和FEch1下调细胞的四种亚细胞组分中表达均定位于细胞质和细胞膜,并且细胞质表达强于细胞膜,细胞核和细胞骨架部分中不表达。结论 Ech1在肝癌组织中的表达明显高于正常肝组织,提示其可能与肝癌的发生、发展有关;体外培养细胞中Ech1表达调控对其亚细胞定位的改变没有影响,Ech1对肿瘤恶性生物学行为的影响可能不是通过其亚细胞定位改变而实现的。
Purpose To study the expression of Eehl in hepatoearcinoma and its clinical significance, and to explore the relationship between subcellular location of Echl and malignant biologic behaviour of hepatocareinoma. Methods hnmunohistochemieal analysis was used to detect Echl expression in the 20 cases of normal liver tissue and the 30 cases of hepatocarcinoma. Subcellular location of Ecbl in Hea-F and FE,.hl -down cell was observed with subcellular protein extraction. Results The expression of Echl in primary hepa- tocarcinoma was increased compared to that of normal liver tissues ( P 〈 0. 05 ) , while there was no significant difference between the expression of Echl with the age, AFP, and with or without liver cirrhosis or hepatitis virus in hepatocarcinoma (P 〉 0. 05). Echl was found expressed almost at the same location although the expression level one is normal and the other is downregulation. Echl expres- sion was found in cytosol, membrane fraction, and its expression was higher in eytosol than other fractions both in Hca-F cell and Eehl downregulated Hca-F cell. Conclusions The expression of Echl in primary hepatocarcinoma was increased, which may indicate that Echl is a critical factor in the development of primary hepatocareinoma, but the subcellular location of Echl has not much contribution to that.
