实验性应激对糖尿病大鼠模型免疫炎症状态影响的研究 被引量:2

Impact of experimental stress on immune inflammatory status in a diabetes rat model

摘要 本文旨在探讨实验性应激对糖尿病大鼠血清IL-2、IL-6、TNF-α水平和脾淋巴细胞活力和凋亡率的影响以及胰岛素和氟西汀对此的干预作用。采用高糖高脂饲养加腹腔少量注射链脲菌素(STZ)诱导糖尿病大鼠,实验大鼠分为正常大鼠对照组、糖尿病大鼠对照组、糖尿病大鼠胰岛素组、糖尿病大鼠氟西汀组,各组大鼠进行为期8周的随机实验性应激,实验第4、8周分别采血测定血清IL-2,IL-6,TNF-α水平,同时检测脾淋巴细胞活力和凋亡率。结果显示糖尿病大鼠应激后IL-6,TNF-α水平明显升高,IL-2水平明显下降;同时脾淋巴细胞活力下降,凋亡率增加,胰岛素或氟西汀治疗能不同程度改善这些变化。 To investigate the impact of experimental stress on serum cytokines and the proliferation and apoptosis of splenic lymphocytes in diabetic rats and the effect of intervention with insulin or fluoxetine.Type 2diabetic rats model were induced by feeding with high sugar and high fat diet and intraperitoneal injection of streptozotocin.The model rats were divided into 3groups,serving respectively as the,Diabetic control group,Diabetic group treated with insulin and Diabetic group treated with fluoxetine.In addition,the normal rats were served as normal control group.All the rats of the 4groups were exposed to multiple stressors for 8weeks.The blood serum was collected from the various groups of rats and was analyzed for levels of serum IL-2,IL-6,TNF-α.Proliferation and apoptosis of splenic lymphocytes were assessed at 4 weeks and 8 weeks.The results showed,compared with thee normal group,the levels of serum IL-6and TNF-αin diabetic rats were significantly increased,the levels of serum IL-2was significantly decreased,the proliferation levels of splenic lymphocytes was significantly decreased and the apoptosis levels of splenic lymphocyte was significantly increased following experimental stress.The above changes were improved to certain extent after being treated with insulin or fluoxetine.
