摘要
目的探讨转移相关基因1(MTA1)和微小染色体维持蛋白2(MCM2)在胃癌组织中的表达分析及其意义。方法选择128例胃癌患者组织作为观察组,癌旁组织作为对照组,采用免疫组化法测定两组组织中MTA1和MCM2蛋白的表达。结果观察组MTA1阳性85例(67.5%),对照组为14例(11.1%),两组比较差异有统计学意义(P〈0.01)。观察组MCM2阳性90例(72.2%),对照组为11例(8.7%),两组比较差异有统计学意义(P〈0.01)。MTA1和MCM2表达尚不能认为与性别、肿瘤分化程度相关(P〉0.05),与肿瘤浸润深度、TNM分期、淋巴结是否转移及是否复发相关(P均〈0.05)。生存分析显示,胃癌患者中MTA1阴性生存时间为56个月,明显长于MTA1阳性的32个月(P〈0.05);MCM2阴性生存时间为54个月,明显长于MTM1阳性的30个月(P〈0.05)。多因素COX回归分析显示,肿瘤的浸润深度、MTA1阳性表达、MCM2阳性表达和淋巴结转移是判断预后的独立预测因素。结论MTA1和MCM2蛋白可以判断胃癌患者的预后。
Objective To study the espression and significance of metastasis-related genes 1 (MTA1) and minichrosome maintenance protein 2 (MCM2) protein in gastric carcinoma. Methods One hundred and twenty-eight gastric carcinoma patients were enrolled into the observation group, while 128 adjacent tissues were enrolled into the control group. MTA1 and MCM2 protein expression were detected through immunohistochemical determination. Results There were 85 cases (67.5%) of MTA1 positive in the observation group and 14 cases ( 11.1% ) in the control group, there was significant difference between the two groups ( P 〈 0.01 ). MCM2 positive were 90 cases (72. 2% ) in the observation group and 11 cases (8.7 % ) in the control group, there was significant difference between the two groups (P 〈 0. 01 ). MTA1 and MCM2 expression had no relationship with gender, tumor differentiation ( P 〉 0.05 ), but had significant relationship with the depth of tumor invasion, TNM stage, lymph node metastasis, and whether recurrence ( P 〈 0. 05 ). Survival analysis showed that the survival time of MTA1-negative gastric cancer was 56 months, which was significantly longer than that of the MTA1-positivegastric cancer (32 months, P 〈 0. 05). The survival time of negatitive MCM2 was 54 months, which was signifi cantly longer than that of the MTMl-positive (30 months, P 〈 0.05 ). Multivariate COX regression analysis showed that depth of tumor invasion, MTA1 expression, MCM2 expression and lymph node metastasis were independent predictor of prognosis. Conclusions MTA1 and MCM2 protein can determine the prognosis of gastric cancer patients.
