黄参多糖提取及其对雏鸡外周血T淋巴细胞增殖的影响

Study on Extraction of Sphallerocarpus Gracilis Polysaccharides(SGP)and the Effect of SGP on Peripheral T Lymphocyte Proliferation in Chicks

为探讨黄参多糖(SGP)的提取纯化和免疫活性作用,采用微波处理法从黄参中提取制备SGP。90℃微波处理90min,过滤,离心,浓缩,用酒精沉淀出粗多糖,冷冻干燥机中冻干,多糖精制,理化性质鉴定。采用MTT法比较不同作用浓度的SGP对鸡外周血T淋巴细胞体外增殖作用的影响。结果表明,SGP得率在微波处理90min时,黄参多糖得率高达5.85%。理化性质鉴定和红外光谱分析,证明提取的SGP纯度较好,不含淀粉、氨基酸或蛋白质。外周血T淋巴细胞增殖试验表明,SGP能够刺激和促进雏鸡淋巴细胞增殖,并且能协同ConA(刀豆素A)刺激雏鸡外周血T淋巴细胞增殖。并且,上述2种刺激作用都具有一定的剂量效应关系,均以SGP终作用浓度在250mg·L-1时效果最佳。SGP具有免疫增强作用。

To study the extraction, purification and immunocompetence of Sphallerocarpus Gracilis polysaccharides （SGP） ,the SGP was extracted with microwave processing for 90 minutes at 90℃, filtrated, centrifuged, condensed, and depositing the crude polysaccharides with alcohol, dried, refined and identification. The effects of SGP on periph- eral T lymphocyte proliferation in chicks was determined by MTT method. The results showed that when the extrac- tion condition of microwave was 90 min, the yield of SGP was 5.85%. The SGP yield was significantly higher with treated by microwave processing and freeze dehydration to obtain the polysaccharide. The results of identification of physiochemical property and infrared spectroscopy proved that the purity of SGP was good, no starch, amino acids or proteins. The data showed that SGP could promote lymphocyte proliferation in chicks. And it also could cooperate with ConA to promote peripheral blood lymphocyte proliferation in chicks. Furhter analysis showed that,both effects related to its dosage,the most effective dose of the SGP concentration is 250 mg · L-1. The SGP can improve the immunocom- petence.
