益气养精小复方对Lewis肺癌荷瘤小鼠免疫功能的影响 被引量:3

Effect of “Yiqi Yangjing Formula”on Immunity Function in Lung Cancer Tumor-bearing Lewis Mice and its Mechanism

摘要 目的:探讨益气养精小复方对Lewis肺癌荷瘤小鼠免疫功能的影响及其作用机制。方法:建立Lewis肺癌细胞C57小鼠皮下移植瘤模型,将造模后的荷瘤小鼠随机分为模型组、中药组、顺铂组和中药+顺铂组。模型组不予药物干预,中药组予益气养精小复方中药灌胃,顺铂组予顺铂腹腔注射,中药+顺铂组予顺铂腹腔注射配合益气养精小复方中药灌胃,连续灌胃给药14 d。比较各组荷瘤小鼠体内抑瘤作用;采用流式细胞术检测荷瘤小鼠外周血T细胞亚群和NK细胞水平;双抗体夹心ELISA法检测荷瘤小鼠血清IFN-γ及IL-10浓度。结果:中药组、顺铂组、中药+顺铂组平均瘤重均显著低于模型组(P<0.05),而该3组之间差异无统计学意义(P>0.05),该3组抑瘤率依次为37.75%、44.59%和45.47%。药物干预后,顺铂组的CD4+T细胞百分比、CD4+/CD8+比值及NK细胞水平均显著低于模型组(P<0.05),其CD8+T细胞百分比显著高于模型组(P<0.05);中药组的CD4+T细胞百分比、CD4+/CD8+比值及NK细胞水平均显著高于顺铂组(P<0.05),而其CD8+T细胞百分比显著低于顺铂组(P<0.05);中药+顺铂组的CD4+T细胞百分比、CD4+/CD8+比值及NK细胞水平均显著高于顺铂组(P<0.05)且低于中药组(P<0.05),其CD8+T细胞百分比显著高于中药组(P<0.05)。中药组与中药+顺铂组血清IFN-γ浓度均显著高于模型组和顺铂组(P<0.05),而IL-10浓度则显著低于模型组与顺铂组(P<0.05)。结论:由生黄芪、黄精、灵芝组成的益气养精小复方能够有效升高小鼠CD4+T细胞百分比、CD4+/CD8+比值与NK细胞水平,正向调节IFN-γ浓度,降低CD8+T细胞百分比与IL-10浓度,进而协同发挥抑制肿瘤生长的作用。 Objective: To investigate the effect of "Yiqi Yangjing Formula" on immunity function in lung cancer tumor- bearing Lewis mice and its mechanism. Methods: Lewis lung cancer cell were implanted subcutaneously in C57 mice to establish transplantation tumor models of Lewis lung cancer. After modeling, the tumor- bearing mice were randomly divided into the model group, Chinese medicine group, cisplatin group and Chinese medicine plus cisplatin group. Except for the model group, others were treated with corresponding measures: the Chinese medicine group was treated with "Yiqi Yangjing Formula" by gavage, and the cisplatin group was injected with cisplatin intraperitoneally, Chinese medicine compound plus cisplatin group was treated with "Yiqi Yangjing Formula" and cisplatin, with the course of 14 days. The anti-tumor effect among the groups were compared in vivo; the peripheral blood T cell subsets and NK cell in tumor-bearing mice were observed by flow cytometry (FCM) ; the concentration of IFN-3, and IL-10 in peripheral blood of tumor-bearing mice were detected by ELISA assay. Results: The tumor weight of the Chinese medicine group, cisplatin group and Chinese medicine plus cisplatin group were obviously lower than that of the model group ( P 〈 0.05 ), and no significant was found among the former three groups ( P 〈 0.05). The tumor weight inhibition rates of the three groups were 37.75% , 44.59% and 45.47% respectively ; after intervention, the percentage of CD4 + , CIM +/CD8 + and NK cell in cisplatin group were obviously lower than that of the model group ( P 〈 0.05 ), and the percentage of CD8 + in cisplatin group was obviously higher than that of the model group ( P 〈 0.05 ). The percentage of CD4 + cells, CIM +/CD8 + ratio and NK cells in the Chinese medicine group were higher than those of the cisplatin group (P 〈 0.05 ), but the percentage of CD8 + was lower than that of the cisplatin group ( P 〈 0.05 ). The percentage of CD4 + , CD4 +/CD8 + and NK cells in Chinese medicine plus cisplatin group were obviously higher than those of the cisplatin group (P 〈 0.05 ), but were lower than those of the Chinese medicine group ( P 〈 0.05 ), and percentage of CD8 + was higher than that of the Chinese medicine group (P 〈 0.05 ). The concentration of serum IFN-y/ of the Chinese medicine group and Chinese medicine plus cisplatin gloup were higher than that of the model group and cisplatin group ( P 〈 0.05 ) , and the IL- 10 concentration was obviously lower than that of the model group and cisplatin group ( P 〈 0.05 ). Conclusion: "Yiqi Yangjing Formula" consists of Radix Astragali seu Hedysari, Polygonati Rhizoma and Ganoderma can improve CD4 +T, CD4 +/CD8 + and NK levels and up-regulate the IFN-yconcentration, reduce CDg + and IL-10 concentration to inhibit the growth of tumor.
