电针对大鼠肛门括约肌损伤后间充质干细胞移植归巢的影响 被引量:2

Effect of Electro-acupuncture on Mesenchymal Stem Cells Transplantation and Homing in Rats after Anal Sphincter Damage

摘要 目的:观察电针对大鼠肛门括约肌损伤后间充质干细胞移植归巢的影响。方法:将72只SD雄性大鼠随机分为空白对照组、假手术组、模型组、电针组、干细胞组和联合组(电针联合干细胞)。采用Zutshi方法复制大鼠肛门括约肌损伤模型;除空白组外,各组分别予相应干预,连续6 d。采用PCR方法检测局部组织基质衍生因子(SDF-1)表达,运用病理技术观察局部组织形态变化。结果:1各干预组SDF-1表达升高,联合组升高最显著(P<0.05);2各干预组新生外括约肌纤维成束,血管管径增大,联合组表现最为明显;3各干预组累积光密度值增高(P<0.05),联合组增高尤为显著(P<0.05)。结论:电针可促进大鼠肛门括约肌损伤后间充质干细胞的移植归巢,有助于肛门括约肌损伤的修复。 Objective:To observed the effect of electro-acupuncture (EA) on bone marrow mesenchymal stem cell (BMSC) transplantation and homing in rats after anal sphincter damage. Methods: Seventy-two SD male rats were randomly divided into six groups: blank control group, sham-operation group, model group, EA group, BMSC group and combined group. The models were replicated by Zutshi method. Except for the blank group, rats in the other groups were intervened by corresponding measures, with the course of 6 days continuously. The expression of SDF-1 was detected by polymerase chain reaction (PCR) and the histomorphological changes were detected by pathological techniques. Results: ①Expression of SDF-1 was increased in all intervention groups, and the combined group displayed high expression ( P 〈 0. 05 ) ; ② Newly born external sphincter fibers was bundled and the diameter of blood vessels were increased, especially in the combined group ; ③ Accumulated optical density was increased in the intervention groups ( P 〈 0.05 ) , especially in the combined group ( P 〈 0. 05 ). Conclusion: EA can promote the transplantation and homing of BMSC and has positive effect to repair the damage of anal sphincter in rats.
