摘要
目的检测长链非编码RNA UCA1在肾癌组织、肾癌细胞系中的表达水平,比较配对组织间、细胞系之间的表达差异,分析UCA1表达水平与患者临床特征之间的关系,探究其在肾癌中的临床意义。方法提取肾癌细胞系及46对配对组织中的总RNA,反转录获得c DNA后通过实时荧光定量PCR方法检测UCA1的表达量,使用统计学软件分析UCA1表达的差异性及其表达水平与患者临床特征之间的相关性。结果 UCA1在肾癌组织中表达水平明显高于配对癌旁正常组织(P<0.001),在肾癌细胞(786-O、ACHN、769P、Caki-2)中表达水平高于人胚肾细胞(293T)(P<0.001)。标本中共有38例(82.609%)肾癌标本UCA1表达上调,癌组织中UCA1表达量与患者临床特征无明显相关性(P>0.05)。结论 UCA1在肾癌组织及细胞系中表达明显上调,提示其和肾癌的发生发展有一定关系。
Objective To detect the expression level of lnc RNA UCA1 in the renal cell carcinoma and cell lines. Then compare the expression in paired tissues and different cell lines, and analyze the relationships between the expression level of UCA1 and clinical characteristics to explore UCA1's clinical significance in renal cell carcinoma. Methods Total RNA was isolated from RCC cell lines and46 paired tissues, and the total RNA was used to get c DNA by performing reverse transcription. Then realtime quantitative PCR(RT-q PCR) was used to detect the expression of UCA1. Statistic analysis was performed to explore the differences of UCA1's expression level and the relationships between the expression of UCA1 and clinical characteristics. Results The expression level of UCA1 was significantly higher in RCC tissues(P〈0.001) than paired normal tissues. And the expression of UCA1 in cell lines(786-O, ACHN, 769 P, Caki-246) was higher than the human embryo kidney cell line(293T)(P〈0.001). 38(82.609%) RCC tissues showed up-regulation in all tissues which were detected. The results showed that there was no relationships between the expression of UCA1 and patients' clinical characteristics.Conclusion RCC tissues and cell lines UCA1 is up-regulated, and this indicates that it may be involved in the tumorigenesis and development of RCC.
