丹参酮ⅡA对糖尿病肾病大鼠肾组织氧化应激的影响 被引量:24

The effects of tanshinone Ⅱ A on oxidative stress of the kidneys of rats with diabetic nephropathy

摘要 目的研究丹参酮ⅡA对糖尿病肾病(DN)大鼠肾组织氧化应激的影响。方法 26只雄性SD大鼠随机分成正常对照(Sham)组、DN组、丹参酮ⅡA组。DN组、丹参酮ⅡA组大鼠经一侧肾切除后腹腔注射链脲佐菌素(STZ)成DN模型,实验8周后分别处死大鼠。测定大鼠体重、血糖、肾功能、尿白蛋白排泄率(UAER),并检测肾组织中的总抗氧化能力(T-AOC)、丙二醛(MDA)和谷胱甘肽-过氧化物酶(GSH-PX)含量及超氧化物歧化酶(SOD)活力。结果 DN组与Sham组比较体重明显减轻,血糖、Bun、Cr、UAER显著增高(P<0.01),肾组织T-AOC和GSH-PX含量下降,SOD活力下降,MDA含量明显增高(P<0.05或P<0.01)。丹参酮ⅡA干预后,上述增高的指标除血糖外均显著改善(P<0.05或P<0.01)。结论 DN大鼠肾脏存在明显氧化应激反应,丹参酮ⅡA具有抗氧化活性,进而起到保护肾脏和延缓DN发生、发展的作用。 Objective To study the effects of tanshinoneⅡ A on oxidative stress of the kidneys of rats with diabetic nephropathy. Methods Twenty-six male SD rats were randomly divided into normal control(Sham) group, diabetic nephropathy(DN) group and diabetic nephropathy group treated with tanshinone Ⅱ A(tanshinone Ⅱ A group). DN group and tanshinone Ⅱ A group rats were intraperitoneal injected with streptozotocin(STZ) after unilateral nephrectomy. The rats were sacrificed after 8 weeks. The body weight, blood glucose, Bun, Cr and UAER were measured in each rat. The levels of total antioxidant capacity(T-AOC), malondialdehyde(MDA), glutathione peroxidase(GSH-PX) and superoxide dismutase(SOD) in renal tissue were determined. Results Compared with the Sham group, the values of blood glucose, Bun, Cr and UAER were significantly increased in the DN group(P〈0.01). The levels of T-AOC,GSH-PX and activity of SOD were decreased and the level of MDA was increased in the DN group compared with the control group(P〈0.05 or P〈0.01). After intervention with tanshinone Ⅱ A, all the indexes measured above were improved except blood glucose(P〈0.05 or P〈0.01). Conclusion This study shows oxidative stress plays an important role in the pathogenesis of diabetic nephropathy in rats.TanshinoneⅡ A has antioxidant effect and tanshinoneⅡ A treatment can protect renal function and delay the development of diabetic nephropathy.
