摘要
目的研究在肝纤维化小鼠肝组织中A20的表达以及炎性反应激活情况。方法通过喂养C57/B6雄性小鼠含有0.1%的3,5-二乙氧基羰基-1,4-二氢-2,4,6-三甲基吡啶(DDC)饲料,构建肝纤维化模型,分别通过Western印迹、PCR检测肝组织中A20、TGF-β、TNF-α、MCP-1、TLR4的表达情况。结果 DDC饲养小鼠肝纤维化明显,小鼠肝组织中A20表达明显升高,A20 mRNA较对照组升高约4.7倍,差异有统计学意义;DDC组小鼠肝内TGF-β、TNF-α、MCP-1、TLR4 mRNA较对照组均升高,分别升高约2.6倍、12.1倍、91.9倍、3.3倍,差异均有统计学意义。结论 A20在肝纤维化小鼠肝组织中的表达增高,A20在肝纤维化的发生发展过程中具有一定的作用。
Objective To investigate the expression of A20 and activation of inflammation in mice with liver fibrosis. Methods The animal model of liver fibrosis were established by feeding food containing 0.1 % of 1 ,4-dihydro-2,4,6-trimethyl-pyridine-3,5-dicarboxylate (DDC)to C57/B6 male mice.The expressions of A20,transforming growth factor-β(TGF-β),tumor necrosis factor-α(TNF-α),monocyte chemoattractant protein-1 (MCP-1)and toll like receptor 4 (TLR4) were measured through western blot and polymerase chain reaction (PCR ).Results The expression of A20 was significantly higher in DDC feeding mice than that in the normal controls,for A20 mRNA increasing approximately 4.7-fold compared with the control group.The expressions of TGF-β,TNF-α,MCP-1 ,TLR4 mRNA were up-regulated by 2.6,12. 1,91.9 and 3.3-fold in DDC feeding group than those in the control group,respectively,which showed statistically significant difference.Conclusion Up-regulation of A20 in liver tissue plays a certain role in the development of liver fibrosis.
