基于表面等离激元共振技术的辐射生物剂量计初探 被引量:2

Study on mice serum CX43 as biodosimetry using surface plasmon resonance technique

摘要 研究小鼠经过不同剂量Co-60γ-射线全身辐照后血清内缝隙连接蛋白Connexin43(CX43)的含量变化,探究其作为生物剂量计的可行性。利用表面等离激元共振(SPR)生物传感器探测小鼠血清内CX43的含量,并采用Western Blot方法对该蛋白进行定性和检验。SPR结果显示,CX43对辐射敏感,0.6 Gy的辐照即可引起小鼠血清内CX43蛋白含量的明显增加。1.2 Gy剂量照射后小鼠血清内的CX43蛋白含量随时间呈先升高后降低的趋势,在3 h左右达到最高峰。此时0-2.4 Gy区间内CX43含量与剂量成线性相关。继续增加剂量后CX43含量反而开始下降。Western Blot结果显示,1.2 Gy照射组净光密度强于0.6 Gy照射组,并且二者的净光密度均明显强于对照组,与SPR结果一致。实验结果表明,基于SPR技术的Connexin43蛋白检测方法有望成为一种新的生物剂量计。 The aim is to investigate the changes of Connexin43(CX43) in mice serum after Co-60 γ irradiation and its practicability as a biodosimetry. The content of CX43 protein in mice serum was detected using surface plasmon resonance(SPR) technique. Western Blot method was also used as reference. Expression of Connexin43 is sensitive to γ radiation. Obvious up-regulation of CX43 expression was observed by γ-rays doses as low as 0.6 Gy. CX43 protein in mice serum first increased and then decreased with the increasing time after mice were irradiated at dose of 1.2 Gy. The highest level of CX43 appeared 3 h after irradiation. At this moment, the content of CX43 showed a linear correlation with the γ-rays doses in the range of 0-2.4 Gy, however, it began to reduce when the dose continued to increase. Western blot results showed that, the fluorescence intensity of the 1.2 Gy group is higher than that of the 0.6 Gy group, and both of them were significantly higher than that of the control group. Detection of Connexin43 protein in mice serum using SPR technique is expected to be a new biodosimetry.
