毛蕊异黄酮对膀胱癌细胞凋亡和PI3K/Akt信号通路的影响 被引量:19

摘要 目的探讨毛蕊异黄酮对膀胱癌细胞增殖、凋亡及PI3K/Akt信号通路的影响。方法采用0、30、60、120μmol/L毛蕊异黄酮处理膀胱癌T24、T739细胞,采用MTT法检测此两种细胞接受上述浓度处理0、24、48和72 h后的细胞增殖情况,流式细胞术检测此两种细胞各浓度处理48 h的细胞凋亡率,Hoechst 33258荧光染色法检测T24细胞各浓度处理48 h的细胞凋亡情况,Real-time PCR检测T24细胞各浓度处理48 h的Bax的mRNA表达水平,Western blot法检测T24细胞各浓度处理48 h的Akt及其磷酸化形式p-Akt的蛋白水平和Bax蛋白水平。结果毛蕊异黄酮抑制膀胱癌T24、T739细胞增殖,并呈剂量和时间依赖性(P<0.05),对T24细胞的抑制增殖效应比T739明显(P<0.05);0、30、60、120μmol/L毛蕊异黄酮作用48 h后T24细胞凋亡率分别为(2.89±1.11)%、(4.11±1.05)%、(16.92±2.67)%、(27.54±2.43)%,而T739细胞凋亡率分别为(3.05±1.42)%、(3.99±1.52)%、(13.01±2.68)%、(19.99±3.31)%,与0μmol/L浓度处理比较,60、120μmol/L处理后明显升高,差异有统计学意义(P<0.05),而且毛蕊异黄酮对T24细胞的促凋亡效应比T739细胞明显(P<0.05);Hoechst 33258荧光染色法显示随药物浓度增高,T24细胞凋亡明显增多;Real-time PCR结果显示T24细胞中Bax的mRNA表达水平在60、120μmol/L处理后均明显高于0μmol/L浓度时的表达水平(P<0.05)。Western blot法检测的Bax蛋白表达水平则在30、60、120μmol/L处理后高于0μmol/L浓度时的表达水平(P<0.05),p-Akt/Akt值则降低(P<0.05)。结论毛蕊异黄酮能抑制膀胱癌T24细胞增殖并诱导其凋亡,其机制可能通过抑制PI3K/Akt通路激活进而促进Bax表达实现。
