Ghrelin对妊娠小鼠子宫内膜蜕膜化的影响 被引量:1

摘要 目的探讨Ghrelin对妊娠小鼠子宫内膜蜕膜化的影响。方法实验将子宫基质细胞分为3组,每组6孔:(1)对照组(只加培养基);(2)Ghrelin组(细胞培养液中加入100 ng/m L重组Ghrelin细胞因子);(3)Ghrelin抗体组(细胞培养液中加入30μg/m L Ghrelin抗体)。通过RT-PCR和免疫组织化学技术,观察Ghrelin及其受体在妊娠小鼠子宫内膜蜕膜化过程中的表达变化;通过子宫基质细胞的分离、培养及宫角注射技术,观察Ghrelin在离体及在体状态下对妊娠小鼠子宫内膜蜕膜化的影响。结果 (1)RT-PCR显示:随着妊娠期和蜕膜化的进展,Ghrelin mRNA表达水平明显升高,至妊娠第8天,其表达水平达到最高。(2)在妊娠第5天,GHS-R蛋白主要表达于腔上皮、腺上皮及胚胎植入位点的蜕膜。至妊娠第6天,其表达水平进一步升高,主要表达于胚胎周围的蜕膜。至妊娠第8天,蜕膜中GHS-R蛋白的表达水平渐进下降。(3)与对照组相比,Ghrelin组子宫基质细胞Hox A10蛋白的表达水平明显升高(P<0.01),而Ghrelin抗体组子宫基质细胞Hox A10蛋白的表达水平明显下降(P<0.01)。(4)与对照组相比,重组Ghrelin对胚胎植入位点没有明显的影响(P>0.05),却明显增加的子宫重量(P<0.05);对于Ghrelin抗体组,能明显抑制胚胎植入位点,且妊娠子宫重量的明显下降(P<0.01)。结论Ghrelin具有明显促进妊娠小鼠子宫内膜蜕膜化的作用。
