摘要 目的:通过紫外线A波段(ultraviolet A,UVA)灯照射快速建立光老化皮肤动物模型,研究光老化皮肤的病理变化及Bax、Bcl-2蛋白表达的变化。方法:应用HE染色及免疫组织化学方法,对经UVA灯照射的大耳白兔光老化皮肤模型进行皮肤组织病理改变及相关凋亡基因蛋白表达的对比观察。结果:大耳白兔皮肤光老化皮肤模型经过UVA灯照射持续24 h后,出现明显褶皱,表皮及真皮结构发生改变,伴有炎性细胞浸润,且相关凋亡基因蛋白Bax表达升高,Bcl-2表达降低。结论:⑴通过UVA灯可以快速、简便、可靠地建立大耳白兔的皮肤光老化模型,为临床进行皮肤光老化的研究提供便利;⑵Bax、Bcl-2的表达可作为衡量光老化皮肤模型造模成功的指标。 Objective To establish the fast dermatoheliosis model in rabbits through exposuring to UVA light,and to investigate the pathological characteristic of skin and expression changes of Bax and Bcl-2 protein. Methods The fast dermatoheliosis model was prepared in rabbits by exposuring to UVA light for 24 h. Then the pathological changes of skin and expression of apoptotic gene such as Bax and Bcl-2 in rabbits model were comparatively analyzed by HE staining and immunohistochemistry. Results The fast dermatoheliosis model was successfully established in rabbits by exposuring to UVA light for24 h,the skin folds occured,the structures of epidermis and dermis were changed,accompanied infiltration of inflammatory cells. Meanwhile,the expression of Bax protein was increased,Bcl-2 protein was decreased. Conclusion The skin dermatoheliosis model could be established by exposuring to UVA light,the method is fast,convenient and reliable,which is helpful for clinical research of skin dermatoheliosis. The expression of Bax,Bcl-2 could be the indicator for evluating whether the model was successfully established.
